Kitab ini bermula dari kegelisahan Imam Jalaluddin al-Suyuthi dalam pengembaraan intelektualnya. Ia heran karena para ulama terdahulu belum ada yang menulis tentang studi ilmu-ilmu al-Qur’an secara komprehensif, sistematis, dan praktis sebagaimana mereka menyusun ilmu-ilmu tentang hadits. Memang, kitab ini sesungguhnya merupakan elaborasi dari kitab yang ditulis oleh Imam Jalaluddin al-Suyuthi sebelumnya, yakni at-Tahbir fi ‘Ulum al-Tafsir. Namun, penjelasan-penjelasan di dalam kitab tersebut sangat ringkas dan hanya memuat 102 topik pembahasan. Sementara itu, di dalam al-Itqan ini, terdapat 80 bab dengan lebih dari 300 topik seputar studi ilmu al-Qur’an. Artinya, untuk mempelajari, memahami, dan kemudian menafsirkan kandungan setiap ayat yang ada di dalam al-Quran, seseorang memerlukan penguasan banyak ragam ilmu. Jilid 2 ini terdiri dari bab 36 sampai bab 40. Bab-bab tersebut di antaranya memuat kata-kata asing dalam al-Qur’an, kosakata serapan (al-mu’arrab), al-wujuh dan an-nazha’ir, makna al-adawat, bahasa selain bahasa Hijaz, dan lain sebagainya.
