"Introduction: At the last step of spermiogenesis, Oko et al. (1993) proposed that unstacked cisternae of the Golgi ribbon organized into a structure previously known as the cytoplasmic droplet, here referred to as the Hermes body. Conserved among all mammalian species, the migration of the Hermes body along the midpiece of sperm in the epididymis is observed to coincide with the initiation of sperm motility. The objective of this study was to compare the most abundant protein constituents of isolated Hermes bodies of epididymal sperm as well as Golgi apparatuses of germ cells in order to provide insight into the formation and function of the Hermes body as well as to ascertain the patterns of expression and differentiation of several Golgi markers during spermatogenesis. Methods: 58 antibodies were selected for immunohistochemical localization (IHC) done on sections of adult male rat testes and epididymides (n=4). Reaction patterns were visualized in all germ cell types across the 14 stages of spermatogenesis, in epididymal sperm throughout the epididymal duct, as well as in all somatic cell types in order to establish each protein's pattern of expression.Results: Each of the 58 antibodies analyzed in this study was investigated as a molecular map of protein expression through the 14 stages of spermatogenesis and for their presence in the Hermes body. Quantitatively, the most abundant Hermes body proteins were those for glucose transport (Glut3) and metabolism (Hexokinase 1), each was localized to the Hermes body by IHC. Actin and myosin were identified to propel the Hermes along the flagellum. In addition, the Hermes body contained proteins characteristic of ER, endosomes, and Golgi apparatus both by proteomics as well as by IHC. In the testis Golgi fraction, the transmembrane 9 family of proteins were prominent and of unknown function along with 200 other proteins. The most abundant protein of this fraction was a novel putative glycosyl transferase (GL54D), an unkown protein but verified by IHC in situ. Golgi proteins conserved between the Hermes and the testis germ cell Golgi apparatus and verified by IHC included the TMED family of transmembrane proteins and COPI/II coatomer proteins. Golgi proteins were also revealed during mitosis, meiosis, acrosome and postacrosomal formation, and Hermes body formation, establishing a protein signature for each during germ cell differentiation. Of the 43 proteins localized to the Hermes body of epididymal sperm, only 31 were maintained down the entire duct, which is explained by the presence of members of the ubiquitin-proteasome system. Discussion: The results of this study reveal that although noted in the Hermes body, Golgi apparatus proteins were not abundant in forming and epididymal Hermes bodies. Hence, the flattened cisternae of the Hermes body may represent a hybrid structure that has evolved to suit the specific needs of the sperm during its maturation.Conclusions: The Hermes body is deduced to compartmentalize glycolysis, cytoskeletal, membrane trafficking, stress response, and protein degradation for the initiation of sperm motility and sperm maturation in the epididymis with its complete protein compendium realized in the step 19 spermatid during Hermes body formation. The Golgi apparatus of germ cells also demonstrates a specific complement of proteins that would serve for the differentiation of germ cells during spermatogenesis." --
