Marine sponges are distributed in the water, from the intertidal zones to thousands of meters deep. They are primitive multicellular invertebrates that live in benthic environments and are bound to solid substrates. Filter feeders, sponges have many microscopic pores on their surface, which allow water to enter and circulate via a network of canals where microbes and organic particles are filtered out and absorbed. Marine fungi are widespread in the oceans and colonize different ecological niches; they are found associated with organisms of all trophic levels and can act as saprobes, symbionts, and parasites. Compared with other marine microorganisms, marine fungus is relatively understudied. Fungi associated with sponges have been discovered to be a promising source of pharmacologically active compounds with unique anticancer, antibacterial, and antiviral properties.

The main focus of this book entitled is to provide an up-to-date coverage of marine sponges and their significance in the current era. This book is an attempt to compile an outline of marine sponge research to date, with specific detail on these bioactive compounds, and their pharmacological and biomedical applications. The book encompasses twenty chapters covering various topics related to Marine Sponges. Initial couple of chapters deal about the worldwide status of marine sponge research, the recent findings regarding dynamics of sponges, and several interesting research areas, that are believed to be deserving of increased attention. Variety of sponges, their toxicology, metagenomics, pharmaceutical significance and their possible applications in biomedicine has been discussed in detail. The second half of this part includes chapters on chemical ecology of marine sponges followed by the discussion on importance of bioeroding sponges in aquaculture systems. The following four chapters of the book deal majorly with the chemical molecules of marine sponges. In the fifth chapter, marine sponge-associated actinobacteria and their pysicochemical properties have been discussed followed by their bioactive potential. The biological application of marine sponges has been presented in later chapters with the classification of biologically active compounds being explored in detail. The second half of the book presents the vast repertoire of secondary metabolites from marine sponges, which include terpenoids, heterocycles, acetylenic compounds, steroids and nucleosides. Further, the bioactive potential of these compounds has also been discussed. One of the constituent chapter elaborates the bioactive alkaloids from marine sponges namely, pyridoacridine, indole, isoquinolene, piperidene, quinolizidine, steroidal and bromotyrosine alkaloids isolated from them. In the next couple of chapters, important sponge polymers and the anticancer effects of marine sponge compounds have been presented. The most interesting aspect of sponge biology is their use in biomedical arena. An effort has been made in this book, to cover the major constituents of sponges and their biomedical potentials. The major portion of sponge body is composed of collagen and silica and used in tissue engineering as scaffold material. This part of the book compiles chapters delineating the isolation of sponge biomaterials including collagen and their use in medical diagnostics. Overall, this book would be an important read for novice and experts in the field of sponge biology.

Biotechnology may be defined as the application of scientific and engineering principles to the processing of materials by biological agents to provide goods and services (Bullet al. , 1982, p. 21) or as any technique that uses living organisms (or parts of organisms) to make or modify products, to improve plants or animals, or to develop microorganisms for specific use (OTC, 1988). The aim of this dissertation was to elaborate the exploration of biologically active secondary metabolites from the marine sponge Cacospongia cf. linteiformis collected from the Bahamas and the soft coral Briareum asbestinum collected from two different sites in Florida State, Boca Raton and Dry Tortugas. In chapter one, a review on previous chemical and biological studies of the marine sponge C. cf. linteiformis and soft coral B. asbestinum is provided. Particular attention is given to spongianolides and briarellins, two important classes of compounds isolated from C. cf. linteiformis and B. asbestinum, respectively, and their structural features and diverse bioactivities. In chapter two, the isolation and relative configuration determination of four epimeric sesterterpenoids, spongianolides E & F (18c, 18d, 19c, 19d) from C. cf. linteiformis collected from the Bahamas are discussed. Thanks to chemical modification (acetylation), diastereomeric 18c&18d and 19c&19d, respectively, were able to be isolated using chromatographic techniques for the first time, and then the relative configurations of 18c, 18d, 19c, 19d were determined based on NOESY NMR experiments. The bioactivity of mixture of compounds 18c, 18d, 19c, 19d were tested and it exhibited inhibition against Schnurri-3 (a regulator of postnatal bone mass). In chapter three, the isolation and structural elucidation of four new compounds, florellins A-D (49-52), from B. asbestinum collected off the coast of Boca Raton, FL are discussed. The molecular structures of these compounds were established by spectroscopic analysis. Compounds 49-52 are the first briarellins containing an acyl group at C-13, while 49 and 50 are the first briarellins possessing acylation at C-15. Florellins A-C (49-51) were screened and found cytotoxic against three human cell lines, BT474, WM266−4 and HEK293. In chapter four, the isolation and structural elucidation of four new compounds, florellins E-H (57-60), from B. asbestinum collected in Dry Tortugas, FL are discussed. The molecular structures of these compounds were established by spectroscopic analysis. Florellins F (58) and H (60) were screened against three human cell lines, BT474, WM266−4 and HEK293, but no cytotoxicity was exhibited. In chapter five, all the experimental procedures are described, including analytical instruments, animal materials, extraction and isolation processes, spectroscopic data and protocols of bioassays.
