There are currently a growing number of laboratories actively studying the mechanism by which various biological membranes are assembled. This area of research is still relatively new to biochemists and molecular biologists, but in view of the rapid progress being made, a review of the field at this time is justified. The present volume focuses on the biogenesis of three related membranes. Mitochondria and chloroplasts are semiautonomous organelles whose biogenesis is carried out partly in the external cytoplasm and partly by the organelles themselves. Both membranes are principally concerned with the energy metabolism of the cell, and this commonality of function is reflected in a considerable degree of similarity in their ul trastructure and enzymatic composition. Although the bacterial cell membrane is a much more diversified structure, it also fulfills the basic energy requirements of the cell, and depending on the organism, this can take the form of photosynthesis or oxidative phosphorylation. The addi tional consideration that prokaryotic organisms may, in fact, be the evolu tionary ancestors of mitochondria and chloroplasts, makes it all the more compelling that those interested in biogenesis be aware of new develop ments in each of these three areas. In organizing this book, I felt that the contributors should summarize and bring up to date their own research and review the literature only in sofar as would be necessary to provide the proper perspective for their work.

Diese Arbeit veranschaulicht am Beispiel der Gerste die Herangehensweise und Durchführung von Versuchen zur Untersuchung chloroplastidärer Proteinkomplexe mit Hilfe der 2D-Gelelektrophorese und deren Auswertung. Die in den Versuchen gewonnenen Daten wurden im zeitlichen Verlauf während der Ergrünung betrachtet. Für den Forscher lassen sich daraus Informationen für die Arbeit mit plastidären Proteinen, deren Biogenese und Kinetik der Proteinassemblierung ableiten. Neben der Anleitung zu Pflanzenzucht und Belichtung werden auch die Isolierung von Mitochondrien, Etioplasten und Chloroplasten detailliert beschrieben. Dabei wurde viel Wert auf die Nachvollziehbarkeit der dargestellten Methoden gelegt. Verschiedene biochemische Trennverfahren werden kurz erläutert und die Gelelektrophorese als eine geeignete Methode zur Auftrennung von Proteinen dargestellt. Es wird auf Varianten der Gelelektrophorese und deren Modifikationen für besondere Anforderungen eingegangen, wie z.B. diskontinuierliche Elektrophoresen und Gradientengele, aber auch denaturierende Elektrophoresen, wie die SDS-Polyacrylamidgelelektrophorese (SDS-PAGE) und die Tricin-SDS-PAGE für eine optimale Auflösung im niedermolekularen Bereich. Besondere Beachtung wird der Blau-Nativen PAGE geschenkt, mit der es gelingt Proteine nativ aufzutrennen, um so vollständige Proteinkomplexe wie Photosysteme und Cytochrome zu unterscheiden und zu identifizieren. Durch Kombination dieser Technik mit der Tricin-SDS-PAGE als zweite Geldimension wird eine 2D-Gelelektrophorese vorgestellt, welche ein Auftrennen einzelner Proteine nach verschiedenen Kriterien erlaubt. Mit der Darstellung der Gele und deren Auswertung konnten Proteine und Proteinkomplexe identifiziert werden und die Veränderung des Proteinmusters während der Ergrünung der Gerste wurde deutlich. Ein anschließender Vergleich der Daten mit der Literatur lässt weitere Interpretationen zu.
