LMO4 is highly expressed in breast epithelial cells and is related to cell proliferation and/or invasion in vivo. Because these cellular features are associated with breast carcinogenesis and since LMO4 is overexpressed in more than 50% of breast cancer cases, we hypothesize that LMO4 may play roles in oncogenesis of breast epithelial cells by regulating proliferation, invasion and/or other cellular features. Using LMO4 over-expression or shRNA expression system in vitro, I found that LMO4 play crucial roles in the regulation of cell proliferation and apoptosis of normal mammary gland epithelial cells or breast cancer cells. Furthermore, I have also observed that deletion of LMO4 impaired the function and development of mammary gland in LMO4 conditional knockout mice, indicating that LMO4 protein is necessary for maintaining the normal development of mice mammary gland. In addition, I demonstrated that the LMO4 can modulate TGF signaling and regulated the proliferative response of epithelial cells to TGF signaling, and thereby linked LMO4 to a conserved signaling pathway that plays important roles in epithelial homeostasis. Under the support of grant, I received excellent training in bioinformatics. By combining previously described functional methods with bioinformatics approaches, we used DNA microarrays to discover LMO4-responsive genes, and identified BMP7 as a key down-stream gene of LMO4. In addition, we also found a significant correlation between LMO4 and BMP7 transcript levels in a large dataset of human breast cancers, providing additional support that BMP7 is a bona fide target gene of LMO4. Finally, we demonstrated that LMO4 binds to HDAC2 and that they are recruited together to the BMP7 promoter. We also suggested a novel mechanism for LMOs; LMO4, Clim2 and HDAC2 are part of a transcriptional complex, and alterations in LMO4 levels can disrupt the complex, leading to decreased HDAC2 recruitment and increased promoter activity.

Chemokines are a superfamily of low molecular weight cytokines that were initially described based on their ability to induce the directed migration of leukocytes to sites of inflammation or injury. In humans, there are approximately 45 chemokines that bind to 19 G-protein-coupled receptors. In addition to mediating cellular migration, chemokines have now been shown to affect many cellular functions including survival, adhesion, invasion, proliferation, and to regulate circulating chemokine levels. Although chemokine receptors were first described on leukocytes, it is now appreciated that chemokine receptors are also expressed by many other cells including endothelial and epithelial cells. Since the first description of chemokine receptors on malignant cells in 2001, an extensive literature has developed describing the expression and function of chemokine receptors in many malignancies. These studies support the initial hypothesis that malignant cells use chemokine receptors to migrate to distant sites of ligand expression and that expression of certain receptors is associated with a poor prognosis. It has also become apparent that malignancies of different tissues may use a diverse profile of chemokine receptors and that the same receptor may mediate metastasis to different sites in tumors of different histological origins. Receptor function may also maintain survival and expansion of the primary tumor.

Nowhere is the explosion in comprehensive genomic testing more evident than in oncology. Multiple consensus guidelines now recommend molecular testing as the standard of care for most metastatic tumors. To aid in the advancement of this rapidly changing field, we intend this Special Issue of JPM to focus on technical developments in the genomic profiling of cancer, detail promising somatic alterations that either are, or have a high likelihood of being, relevant in the near future, and to address issues related to the pricing and value of these tests. The last few years have seen the cost of molecular testing decrease by orders of magnitude. In 2018, we saw the first "site-agnostic" drug approvals in cancer (for microsatellite unstable cancer (PD-1 inhibitors) and NTRK-fusions (TRK inhibitors)). Research on targetable mutations, determination of genetic "signatures" that can use multiple individual genes/pathways, development of targeted therapy, and insight into the value of new technology remains at the cutting edge of research in this field. We are soliciting papers that present new technologies to assess predictive biomarkers in cancer, original research (pre-clinical or clinical) that demonstrates promise for particular targeted therapies in cancer, and articles that explore the clinical and financial impacts of this paradigmatic shift in cancer diagnostics and treatment.
