"The remarkable and unique mechanical properties of natural structural designs have always been a source of inspiration for scientists to enhance material design. The integration of bioinspired design with topology optimization promises unprecedented possibilities in terms of improving the properties and functionalities of products. Recently, the advancement in additive manufacturing has enabled the sustainable and accurate fabrication of complex designs that were not possible a decade ago, which opened the door to implement and develop material design optimization methods. Aiming to understand and enhance design optimization methods towards the advancement of sustainable product design, the present dissertation systematically examined various optimization techniques and their implementation in real-life applications with a focus on a bioinspired approach. The dissertation involves design optimization through both analytical modeling and finite element analysis (FEA). Uniaxial tension and compression experiments were conducted on additively manufactured specimens to validate the results obtained from the different investigated design optimization models. The dissertation is manuscript-based containing three inherently connected articles (see the first page in chapters 3-5 for the interconnectivities). Chapters 3 covers design optimization through analytical modeling with a focus on optimizing the design of circumferential clasps in additively manufactured removable partial dentures. Chapter 4 presents bioinspired shape optimization with mesh adaptive direct search to improve strength and stiffness of metal/polymer interfaces in additively manufactured dental prostheses. Chapter 5 introduces an optimization framework for bioinspired topological design coupled with probability distribution models to generate the initial guess. Through this framework, bioinspired and optimized cellular structures could be designed for various biomedical and engineering applications. The aforementioned studies offer new insights to leverage the potential of additive manufacturing and optimize the mechanical design of structures and interfaces"--

Biological systems are composed of complex materials which are responsible for performing various functions, such as providing structural support, mobility, functional adaptation to the environment, damage repair, and self-healing. These complex materials display excellent mechanical properties and can rapidly adapt to external stimuli. Thus, nature inspires in terms of source materials, functions, and designs to develop new-generation structural and functional materials. Polymers (natural or synthetic) are excellent sources of developing materials to mimic the functions of soft segments in biological systems. This dissertation focuses on synthesizing and characterizing two different materials with tunable structures and properties: complexes from natural polysaccharides or polyelectrolytes and bioinspired hydrogels from synthetic polymers. Oppositely charged polyelectrolytes can form polyelectrolyte complexes (PECs) due to the electrostatic interactions. The structure and properties of PECs can be tuned by varying the salt concentration, as the addition of salt can facilitate associative phase separation. PECs were prepared from two biopolymers, positively charged chitosan and negatively charged alginate. Rheological experiments for the complexes displayed a tunable shear modulus with changing salt concentrations. The microstructural study conducted using small-angle X-ray scattering provided insights regarding the length scales of these complexes, and the results follow the observed rheological and phase behavior. Elastic biopolymers such as resilin display remarkable mechanical properties, including high stretchability and resilience, which many species exploit in nature for mechanical energy storage to facilitate their movement. Such properties of resilin have been attributed to the balanced combination of hydrophilic and hydrophobic segments present in the chain. In this work, we synthesized hydrogels with hydrophilic and hydrophobic components to mimic the properties of resilin. With this system, we determined the tensile, retraction (ability to revert to the original state after stretching), and swelling properties when (i) the concentration of the hydrophobic polymer was varied and (ii) additional hydrophobic components were included. The stretchability, stiffness, and strength of the gels varied as the compositions were altered. The fundamental understanding of the structure-property-function relationship for materials presented in this work provides insights into engineering materials for applications such as tissue engineering, drug delivery, wound healing, artificial muscles, soft robotics, and power amplification.

Nature has developed advanced materials after billions of years of evolution in an exquisite way, giving rise to elaborate materials with optimized designs, architectures, properties, and functionalities. These functionalities emerge from the directed assembly of biomolecules from the molecular level up to the length scales relevant to living cells, cellular tissue, and entire organisms. Inspired by these bottom-up assembly processes, I have developed materials that self-assemble into active substrates mimicking the actuation found in living muscle fibers. The resulting bioinspired materials display properties that mimic endogenous viscoelastic properties found in collagen and, consequently, push muscle cellular differentiation faster than the state-of-the-art elastic scaffolds. During my PhD, I developed a nonwoven CLC electrospun scaffold by dispersing three cholesteryl ester-based mesogens within polycaprolactone (PCL). We tuned the ratio of our mesogens so that the CLC would be in the mesophase at the cell culture incubator temperature of 37 °C. In these scaffolds, the PCL polymer provided an elastic bulk matrix while the homogeneously dispersed CLC established a viscoelastic fluidlike interface. Increasing the CLC concentration within the composites enhanced myoblast adhesion strength promoted myofibril formation in vitro with mouse myoblast cell lines. Cholesteryl ester liquid crystals (CLCs) intrinsic ability to organize into supramolecular helicoidal structures on the mesoscale allows cell-helix interaction to create dynamic biomaterials. Inspired by the shoaling behavior of fish, I devised a method to deploy the CLC-helix to orchestrate and to template the three-dimensional aggregation of anchorage-dependent cells lines. The 3D cellular aggregates are rapidly produced off the dynamic scaffold providing a platform to manufacture cost-effective organoids for precision medicine and cellular agriculture. As scientists, it is our calling to provide solutions to protect society. During my PhD, we were faced with the Coronavirus (COVID-19) pandemic. We developed a self-cleaning fabric ambient environment sunlight provides enough energy to stimulate the band gap of embedded 3D anisotropic nanoparticles fiber matrix, mixing the dimensionality of the nanomaterial mentioned gives rise to rose-thorn-like protrusions a natural defense plant morphology seen in nature. Herpes simplex virus (HSV) plagues billions of humans with infections globally. We have developed and demonstrated rose-thorn-inspired antiviral fibrous arrays by electrospinning a composite of polycaprolactone (PCL) polymer with a dispersion of anisotropic zinc oxide tetrapod nanoparticles (ZOTeN). This rose-thorn-mimicking material enables physical and chemical protection. Under blue-light stimulation, ZOTeN photocatalyzes the production of hydrogen peroxide for an accessible disinfection and sterilizing mechanism to prolong materials usage. The fibrous material has dose-dependent antiviral properties against both HSV-1 and HSV-2. The engineered mats can potentially be used for manufacturing antiviral garments, face coverings, and bandages.
