"The aim of the project was to develop an improved protocol that increased the yield of DNA isolated from bone. Bone is resistant to decay (Ye, 2004), and after disasters or other mayhem bone fragments may be all that is left to identify those who died. One of the first successful extractions of DNA from bone used ancient samples that were between 300 and 5,500 years old (Hagelberg, 1989). Recently, collagen fibers that were preserved in fossilized bone from both a 160,000 to 600,000 year old mastadon, and a 68 million year old dinosaur were successfully analyzed (Asara, 2007: and Schweitzer, 2007). Findings from one study suggested that the bone mineral had undergone little if any change from its living state since the 68 million years ago when it died (Schweitzer, 2007). The most widely used protocol for extracting DNA from bone can be found in the Forensic Science Handbook: Volume II 2nd ed. (Saferstein, 2005). The procedure often fails to yield usable DNA, especially whensamples are small. The goal of this research was to increase the efficiency of DNA extraction to improve reliability of bone use for forensics and other purposes. In addition to the extraction of DNA from bone, a novel procedure for the extraction of DNA from bird feathers was developed. This source of DNA is useful for the sexmg of birds from a single down feather by applying common molecular technologies. The down feathers are easily collected and collection poses little physical harm to the bird. This is particularly useful in light of the fact that over fifty percent of avian species appear to be identical, sexually uniform in their morphology (Griffiths, 1996). In avian species the female is the heterogametic sex (ZW), while the males are homogametic (ZZ), a motif opposite of humans. The chromo- helicase-DNA-binding (CHD) genes, located on the sex chromosomes, enable molecular sex determination (Griffiths, 1996). The DNA of female birds has both the CHD-W and CHD-Z genes on their W and Z chromosomes, respectively. Amplification of both sequences during PCR generates DNA distinguishable from male birds because males contain two copies of the CHD-Z gene and no CHD-W. When the copies of the genes are run during gel electrophoresis, female samples are visually identified as having two bands, while the males have only one. By utilizing only down feathers, the bird sexing assay becomes completely non-invasive and poses little physical distress to the bird"--Document.
