In order to compare the results of the analysis from this point of view it is necessary to start with the contents of the first sample analyzed and to reduce the other in such proportions that their contents may accurately correspond to those of the first sample. A calculation of this kind may be based upon the contents in sodium chloride, which exists in the fermenting mass in a large quantity without being subjected to any conversion, the quantitative determination of which affords great security, because it can be carried out with ease and accuracy. Hence we reduced the contents of the 2nd, 3rd and 4th samples to the proportion of common salt contained in the first sample (21.43 parts); for example, the amount of glucose in the 2nd sample, corresponding to the substance of the first sample is found by the equation: 23.35: 15.28=21.43: X; X=i4.02. In this manner the following figures were obtained: --It appears herefrom in general that the fermentation was accomplished within the last ioo days of the process rather uniformly, its intensity showing no diminution and the losses of dry and organic matter exhibiting the almost regular proportion i: 2: 3 for the intervals of 35: 70: 100 days. This indicates that at the end of the 5 months of the whole process the fermentation was not yet complete, but would have probably continued still longer. The destruction concerns most largely the carbohydrates (starch, dextrin, glucose, etc.), the majority of which, probably after saccharification are oxydized. The quantity (20.08 parts) decomposed of them does not, however, cover the whole loss of organic matter (22.77 parts), 2.69 parts at least being left unaccounted for, which...

Excerpt: ... development of new rootlets takes place with more difficulty, and accordingly, their absorptive power must remain comparatively stationary and the recovery will consequently be slower. For the same reason, the disease appears oftener after cutting in late summer or autumn, while it is less frequent when the plants are cut earlier. The development of new rootlets and the consequent recovery after cutting, must be much more difficult in autumn. The young rootlets must once lose their activity after cutting, and the entire root-system will become gradually inefficient, unless the development of the new rootlets is exceptionally good; and after successive cuttings, the small roots must entirely die off. So it is quite natural that we always find decayed roots when the disease is much advanced. But in the earlier stages of the disease, we find only small rootlets dead. The roots and leaves are closely correlated to each other, a bad condition in the one accelerating the degeneration of the other; and the primary cause of the disease must be sought in the practice of cutting in the growing period. We see Plates XIX-XXVI that the quantity of reserve nitrogen in the roots of Takasuke, Tsuruta etc. is not restored to its norm even in December. This evidently shows a gradual wearing of the plants, and after two or three years, they must become diseased unless they are kept from cutting. In Japan occurs in commerce a food called " Koiinyakit," prepared from the root of Conopliallus Konyakit, by treating with dilute milk of lime, and resembling stirch paste, which consists almost exclusively of mannan.3 For my experiments I prepared a much more diluted product dissolving 3% of the refined dry product of the said root in a hot solution containing % of pepton and O.l% of magnesium sulphate and dipotassiumpliospliate. This solution gave on cooling a transparent jelly resembling the agar jelly, used for bacteriological cultures. 1 This calamity was easily avoided by antiseptic means, which the writer proposed. 2 Tsuji. It was only Bacillus inesentericus vulgatns of the sixteen species tested that liquefied mannan in two days, the same microbe which also can saccharify starch. But according to my observation it does not saccharify araban. The above named microbe cultures were observed, for several weeks further, but it was only Bacillus prodigiosus that showed a weak action on mannan within that time. The careful examination of the spoiled mucilage mentioned above convinced the writer, that the loss of viscosity was caused by Bacillus mesentcrictts inilgatus. Further no trace of development was noticed, when this yeast was transfered into well aerated nourishing solutions. An observation was, however, made during all these experiments which may explain the contradictory statements of those observers who claimed that the fermenting power is destroyed by a temperature of 40. I have found that yeast exposed to heat for some time never showed within such short time fermentation in Pasteur s solution as the control sample; sometimes the retardation amounted to 12 hours, sometimes to several days. In some of the cases mentioned, viz., when yeast was heated for 27 or 29 minutes to 5OC. a weak fermentation was afterwards observed with these samples but after a few days it gradually stopped again, long before all the sugar of the solution was fermented. But those samples of yeast that had only been exposed for 25 minutes to 5OC., still preserved all their vital powers and were observed to develop well, when inoculated on nourishing gelatine, potato, or in i% sugar and pepton solution. I, therefore, infer that for 25 minutes a temperature of 50 forms the limit beyond which not only the fermenting power but also all other life functions are destroyed. It was my further object to test the influence of various compounds upon the resisting power of the yeast-cells against heat. The experiments were carried on in the same way as described above, but the yeast was here suspended in different solutions instead of in distilled water, while the liquid was heated.
