Cell death is one of the fundamental processes by which normal development is modulated, and the importance of both necrosis and apoptosis in a number of pathologies has generated intense interest from researchers in many fields. This timely book covers both the proteins that are produced by dying cells and the proteins that signal cells to initiate cell death. Cell Death Proteins provides an overview of the explosive interest in cellular death. Six review papers, written by researchers at the forefront of this rapidly moving field, focus on proteins that promote, signal, and inhibit cell death. Major players involved in the cell death cascade and its controls are covered, including cell cycle checkpoints, the function of interleukin-1J converting enzyme, the role of IGF-I receptor, the Bcl-2 family of proteins, viral inhibitors of apoptosis, and p53-dependent apoptosis.

Beginning from centuries of anecdotal descriptions of cell death, such as those on the development of the midwife toad in 1842 by Carl Vogt, to modern-day investigations of cell death as a biological discipline, it has become accepted that cell death in multicellular organisms is a normal part of life. This book provides a comprehensive view of cell death, from its mechanisms of initiation and execution, to its implication in human disease and therapy. Physiological cell death plays critical roles in almost all aspects of biology, and the book details its roles in lymphocyte homeostasis, neuronal function, metabolism, and the DNA damage response. When physiological cell death goes awry, diseases can arise, and cancer is presented as a central paradigm for the consequences of derangements in the interplay between cell survival and cell death. At the same time, the potential promise of targeted therapies aimed at interdicting cell death machineries are also discussed extensively. The molecular mechanisms that underlie apoptotic cell death are illustrated from the perspectives of both the intrinsic, mitochondrial apoptotic pathway and the extrinsic, death receptor pathway. Key players in these pathways, such as the Bcl2 family proteins, cytochrome c, Apaf-1, caspases, death receptor adapter proteins, and inhibitor of apoptosis proteins, are presented from both functional and structural angles. Until only a few years ago, programmed cell death has been considered essentially synonymous with apoptosis. However, we now know that programmed cell death can also take other forms such as necrosis or necroptosis, and to this end, the mechanisms that underlie programmed necrosis in development and host defense are illustrated. The past twenty plus years have seen an incredible growth of research in cell death, with one breakthrough after another, and the legacy still goes on with constant new surprises and findings. Long live cell death!

Bcl 2 Protein Family written by Claudio Hetz and has been published by Springer Science & Business Media this book supported file pdf, txt, epub, kindle and other format this book has been release on 2011-01-12 with Medical categories.

In this book, scientists pioneering the field have compiled a series of focused chapters to highlight the relevance of the BCL‐2 family of proteins in apoptosis, physiology and disease. An important focus of this volume is considering the potential TH ERA PEUT IC benefits of targeting apoptosis pathways in the context of human disease. Readers interested in understanding how a cell handles stress and the consequences of dysregulation of this process for human disease will find this book very valuable. It attempts to describe a fascinating area of research where physiology and biomedicine converge at different levels, revealing a trip from the molecular regulation of apoptosis to the impact of this process to the physiology of a whole organism.

Proteases In Apoptosis Pathways Protocols And Translational Advances written by Kakoli Bose and has been published by Springer this book supported file pdf, txt, epub, kindle and other format this book has been release on 2015-08-06 with Science categories.

This book provides a comprehensive overview of the proteases involved in programmed cell death. It presents a focused yet extensive discussion on proteolytic enzymes such as caspases, HtrAs, granzymes, calpains and cathepsins as well as laboratory protocols related to enzymology and apoptosis. Mouse model systems and non-invasive imaging techniques in apoptosis-related diseases such as cancer and neurodegeneration are also covered in this book. While slowly unravelling the complexities of apoptosis in chapter one, the next three chapters individually elaborate on different classes of proteases that play key roles in the initiation, progression and execution of programmed cell death. The last two chapters complete this discussion by describing different laboratory methodologies and therapeutic advances involving apoptotic proteases. Protocols portraying in vitro and ex vivo colorimetric and fluorescence-based enzyme kinetic studies as well as cell death assays are explained in the fifth chapter. Preclinical in vivo models and non-invasive imaging in apoptosis to understand the complexities of disease progression and their contribution toward therapeutics is recounted in the last chapter. The book spans topics related to both fundamental and applied biology. It would therefore be equally appealing and informative to scientists working in the field of apoptosis and those who are investigating mechanisms of proteases and enzymes in general. The protocols would certainly benefit both graduate and undergraduate students working in the related fields and provide useful leads for drug design to translational biologists involved in neurodegeneration and cancer research.
