Our knowledge of cholinergic synaptic transmission in the peripheral nervous system (PNS) has expanded enormously since the early 1930's, at which time O. Loewi discovered chemical neurotransmission through acetylcholine (ACh) and the pharmacological actions of ACh were described by H. Dale and his colleagues. Description of ACh's actions and receptors in various parts of the brain was followed by a detailed analysis of ACh's synthesis, release mechanism, removal from the synaptic cleft, modes of agonist-receptor interactions, properties of regulated conductances and of the pre-and postsynaptic modulation of cholinergic synapses. Our knowledge has been increasingly consolidat ing, leading us to the recent clarification of the structure of the periph eral nicotinic receptor-ion channel and its functional properties. It is appropriate to maintain the claim that the fundamentals of our under standing of synaptic transmission come from studies on cholinergic synapses in the PNS. Contrastingly, views held on the role of cholinergic synapses in the mammalian central nervous system (CNS) are extremely controversial, although it has been clear for some time that ACh is present in the brain. Illustrating this, no unanimous view is held on the role of nicotinic receptors in the CNS. There is one notable exception to this confusion: Studies begun by J. C. Eccles and associates in the 1960's and completed by D. R. Curtis and R. W.

Synaptic transmission plays a central role in the nervous system as the mechanism that allows for chemical and electrical communication between cells and thus connects discrete elements into the functioning whole. This is a broad account of anatomical, biochemical, embryological, medical, pathological, pharmacological, and physiological studies on synaptic transmission during the hundred years beginning in 1890. During this century, the process of synaptic transmission came to be recognized not only as the most fundamental neurophysiological process, but also as a seat of pathological changes, and as the predominant site of action for drugs used to treat a wide range of psychiatric and neurological disorders. At the same time, research from these various disciplines was transformed into a new and unifying field, neuroscience. The course of these investigations reveals ingenious experiments, powerful new techniques, and imaginative insights. The author describes broadly who did what, when, where, and how (and, in cases where it is apparent, why) and uses experimental results and interpretations to display the evolutionary course to our current understanding of how nerve cells communicate: the basic principle of neural functioning. The book will be of interest to basic and clinical neuroscientists, pharmacologists, and physiologists, to historians and philosophers of the life sciences and medicine, and to their respective students.

Even if the “weapons of mass destruction” (WMD) and, among them, stocks of organoph- phorus (OP) agents (also referred to as war gases and nerve gases) were not found in Iraq following the US-Iraq war, the relative ease with which these substances can be made from harmless precursors and the low cost of their manufacture will continue to fascinate pow- hungry, ruthless dictators, as well as multinational and international terrorists, particularly as the close relationship between the OP agents and useful insecticides makes it easy to disguise the importation and purchase of small amounts of the precursors. Indeed, the use by Saddam Hussein of a nerve gas against the Kurds and his possible employment of the OP agents during his war with Iran, and the Sarin attack in the Tokyo underground by an extremist religious set magnetized the world with respect to the OP drugs. As these drugs exert their toxicity via their cholinergic action on the nervous, particularly central nervous, system, it is no wonder that the research in the cholinergic ? eld attracts, and merits, our intense attention. These considerations underlie the signi? cance of this book, as Alex Karczmar devotes an entire chapter of Exploring the Vertebrate Central Cholinergic Nervous System to anticholinest- ases (antiChEs), and as he is an acknowledged expert in the ? eld of cholinergic toxicity as well as a consultant to the Surgeon General of the U. S. Army.

Pharmacology of Central Synapses deals with the pharmacology of the central nervous system and the mode of action of neurotropic drugs based on their influence on synaptic transmission. The neuronal structure of the nervous system, the structure of the synapses, and the phenomena of electrochemical transmission are discussed in this context. The main propositions of the synaptic theory of the action of neurotropic drugs, based on morphological, physiological, and biochemical findings, form the conceptual basis of this book. Comprised of eight chapters, this book examines the effect of neurotropic drugs on the synaptic transmission in reflex, projection, and association (commissural) tracts. The specific effects of various narcotics, neuroleptics, tranquillizers, analgesics, antidepressants, and stimulants on synaptic transmission in various structures of the central nervous system and at its different levels are described. This text also considers the differences in the sensitivity to such drugs of various synapses and their influence on the selectivity of drug action. This book explains the influence of neurotropic drugs on such aspects of nervous activity as impulse summation, after-discharge and functional lability, central inhibition, and the neurochemical mechanisms, particularly those involving monoaminergic, cholinergic, serotoninergic, and GABA-ergic components of synaptic transmission. The final chapter focuses on the effect of psychotropic drugs on behavior. This book will be helpful to scientists representing all the major areas of pharmacology, including clinical pharmacology and toxicology, as well as to internists, psychiatrists, neurologists, and anesthesiologists.

Chemical Transmission of Nerve Impulses: A Historical Sketch is a translation of the French edition """"Les Transmissions chimiques de l'influx nerveux"""" published by Gauthier-Villars in Paris. Organized into 12 chapters, this book begins with a discussion on the definition and importance of the phenomenon of chemical transmission, with chapters specifically devoted to both cholinergic and adrenergic ones. This text then elucidates the general organization of the nervous system in vertebrates. The opposition to the theory of chemical transmission and the state of comparative physiology with ...
