Cytokines such as transforming growth factor β (TGFβ) and tumour necrosis factor α (TNFα) have also been implicated in regulation of osteoclastogenesis and osteoclast function indirectly. These two cytokines exert their effects by acting upon OPG, RANK, RANKL, or M-CSF. From the literature TGFβ was presented with conflicting in vivo and in vitro effects on bone resorption. Our results support the inhibitory effect of TGFβ and demonstrate TNFα to be a stimulator of bone resorption. When it comes to immunity, interferons (IFNs) play a crucial role and they also play an important role in osteoclast biology. To study the effects of type 1 IFNs in vivo we generated Ifnar1 and Ifnar2 deficient mice. These mice were shown to be null for conventional IFN-induced Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling. Our studies on Ifnar1 and Ifnar2 knockout mice demonstrated enhanced generation of osteoclasts in vitro. Consequently the number of multinucleated TRAP positive cells was increased and as expected mice developed osteoporotic phenotype, characterised by reduced bone density. Many other cytokines, proteins and various other factors play an important role in the proliferative phase of osteoclast precursors and/or in the terminal differentiation phase and/or in the bone resorbing activity of fully matured osteoclasts. It is therefore of uttermost importance to account for the fact that osteoclast biology and bone metabolism (as a whole) are regulated via rather complex series/cycles of events that are accomplished at different levels and stages through different pathways.
