Headstarting is a conservation strategy that assumes raising hatchling turtles to larger body sizes increases their survivorship compared to wild non-headstarted turtles. This increased survivorship should increase population growth rate relative to wild recruitment. There are, however, few published results of long-term population recovery using headstarting. The lack of demographic assessment of population recovery has led to an overall lack of quantitative assessment of the effectiveness of headstarting as a conservation action. Headstarting needs to be efficient and effective as a poorly executed headstarting project can result in species extinction given it is often used with critically endangered species. We released 3 cohorts of headstarted Wood Turtles (Glyptemys insculpta) with varying degrees of headstarting to determine if headstarting increases survivorship. I showed that headstarting turtles to a larger body size confers a survival advantage, and this survival advantage should increase population growth rate relative to wild recruitment. I then quantitatively assessed the effectiveness of a 15-year Wood Turtle headstarting program by modeling population-specific demographic parameters to evaluate recovery efforts, and determine the next phase of recovery. I found some evidence of population recovery, but also identified challenges and make several management recommendations that should enhance the success of the headstarting program. Overall, I have provided support for headstarting as an effective conservation strategy, with the caveat that all headstarting projects must be paired with management plans that maintain high adult and juvenile survival.

Thermal ecology studies of ectotherms, like turtles, have typically focused on a species' thermal preferences and tolerances, or on thermoregulation site selections; only recently have landscape-scale thermal ecology studies been performed. I examined the spatial and nesting ecology of wood turtles in Sudbury District of Ontario, Canada, in a thermal context. I also measured the thermal impacts of natural resource extraction on wood turtle habitat. Wood turtles (Glyptemys insculpta) cover a wide variety of terrestrial and aquatic habitats during their annual cycle, making them ideal for thermo-spatial studies. I tracked movements and thermal use of 15 radio-tagged adult turtles during the active season, comparing their selections to temperature monitoring stations spread in an array across the study area, to determine if the turtles are navigating a thermal landscape. Temperature had minimal influence on home range-scale movements, but possibly influenced movements at a smaller spatial scale. I compared the thermal landscape (using thermal imagery), soil moisture, and grain size distribution of 3 nesting beaches to determine the strongest predictor of nest-searching behaviour. Temperature range appeared to be an important cue, but females were apparently using a suite of cues to select their nest sites. I mapped the thermal landscapes of six sites: two relatively undisturbed wood turtle habitat sites, two recently-harvested forestry sites, and two active gravel pits, to find the effects of resource harvesting on wood turtle habitat. The undisturbed sites were cooler and less variable than the disturbed sites, and provided higher-quality thermal habitat. My results support the findings of previous studies: that temperature is a stronger driver of turtle behaviour at the micro-habitat scale than the home range scale, and that soil temperature co-varies with soil structural variables at the micro-habitat scale. The data from the habitat mapping provide useful information for conservation efforts when mitigating or rehabilitating wood turtle habitat.

Worldwide turtle and tortoise populations are declining, including the semi-aquatic wood turtle (Glyptemys insculpta) of eastern North America. With a limited survey season, a more efficient method of detection was needed to monitor this cryptic species. DNA barcoding with environmental DNA (eDNA) has been shown to be an effective method of monitoring cryptic, rare, and threatened species. Water samples were collected in October 2017 and 2018 from an upstream and downstream site on fourteen rivers in New Brunswick, Canada. In our eDNA lab, the samples were filtered, extracted and, with the use of species-specific primers and probe, amplified using quantitative polymerase chain reaction (qPCR). With the use of qPCR, wood turtle eDNA was detected in five of six rivers known to inhabit the species. This method is a promising tool that can be used for the conservation and monitoring of G. insculpta.
