Johann Wolfgang von Goethe was a German writer and statesman. His body of work includes epic and lyric poetry written in a variety of metres and styles; prose and verse dramas; memoirs; an autobiography; literary and aesthetic criticism; treatises on botany, anatomy, and colour; and four novels. In addition, numerous literary and scientific fragments, more than 10,000 letters, and nearly 3,000 drawings by him are extant. A literary celebrity by the age of 25, Goethe was ennobled by the Duke of Saxe-Weimar, Karl August in 1782 after first taking up residence there in November 1775 following the success of his first novel, The Sorrows of Young Werther. He was an early participant in the Sturm und Drang literary movement. During his first ten years in Weimar, Goethe served as a member of the Duke's privy council, sat on the war and highway commissions, oversaw the reopening of silver mines in nearby Ilmenau, and implemented a series of administrative reforms at the University of Jena. He also contributed to the planning of Weimar's botanical park and the rebuilding of its Ducal Palace, which in 1998 were together designated a UNESCO World Heritage Site. Arthur Schopenhauer cited Wilhelm Meister's Apprenticeship as one of the four greatest novels ever written, along with Tristram Shandy, La Nouvelle Heloise, and Don Quixote, and Ralph Waldo Emerson selected Goethe as one of six "representative men" in his work of the same name, along with Plato, Napoleon, and William Shakespeare. Goethe's comments and observations form the basis of several biographical works, most notably Johann Peter Eckermann's Conversations with Goethe. There are frequent references to Goethe's writings throughout the works of G. W. F. Hegel, Arthur Schopenhauer, Friedrich Nietzsche, Hermann Hesse, Thomas Mann, Sigmund Freud, and Carl Jung. Goethe's poems were set to music throughout the eighteenth and nineteenth centuries by a number of composers, including Wolfgang Amadeus Mozart, Ludwig van Beethoven, Franz Schubert, Robert Schumann, Johannes Brahms, Charles Gounod, Richard Wagner, Hugo Wolf, Felix Mendelssohn, and Gustav Mahler.

The effort to sequence the human genome is now moving toward a c- clusion. As all of the protein coding sequences are described, an increasing emphasis will be placed on understanding gene function and regulation. One important aspect of this analysis is the study of how transcription factors re- late transcriptional initiation by RNA polymerase II, which is responsible for transcribing nuclear genes encoding messenger RNAs. The initiation of Class II transcription is dependent upon transcription factors binding to DNA e- ments that include the core or basal promoter elements, proximal promoter elements, and distal enhancer elements. General initiation factors are involved in positioning RNA polymerase II on the core promoter, but the complex - teraction of these proteins and transcriptional activators binding to DNA e- ments outside the core promoter regulate the rate of transcriptional initiation. This initiation process appears to be a crucial step in the modulation of mRNA levels in response to developmental and environmental signals. Transcription Factor Protocols provides step-by-step procedures for key techniques that have been developed to study DNA sequences and the protein factors that regulate the transcription of protein encoding genes. This book is designed as a course in the phonetic transcription of normal and disordered speech. What differentiates this book from existing approaches to phonetic transcription and conversational analysis is that it concentrates on linking together layers of detail to result in a complete record for the entire range of transcribable behaviors. Müller's book represents the first attempt to amalgamate differing methods to give phoneticians and clinicians a transcriptional tool kit, thus allowing them to generate a rich description of their data. This approach results in a variety of layers of transcription, all or some of which are available to the clinician or researcher faced with the task of transcribing speech. The layers include a base, orthographic layer; segmental and suprasegmental phonetic layers; a gaze and gesture layer; a layer for marking aspects of discourse (e.g., overlap); and finally, a layer for highlighting behaviors of specific clinical interest (e.g., stuttering behavior). This book clearly lays out the various layers of transcription in this approach, illustrating them with normal and clinical data as well as exercises for the reader. Each chapter in the book addresses a different layer of transcription, with a final chapter illustrating how to bring the layers together. Worked examples accompany each chapter, and appendices provide a quick reference to symbols and transcription conventions.
