Abstract: Nine papers developed from a 1981 international workshop summarize current knowledge of the role of proteins (and amino acids) in lipid metabolism and atherosclerosis for nutritionists, clinicians, and other health professionals. The topics range from studies on lipid metabolism and experimental atherosclerosis in animals (rats, rabbits, swine), to effects of proteins on human serum lipids and lipoproteins. Other papers discuss the effects of soy protein and casein, influences of dietary protein on serum cholesterol, and the immunological aspects of atherosclerosis. The results of these studies indicate that dietary protein can make a significant contribution to atherosclerosis, that soy protein produces lower serum cholesterol levels than casein (the dynamics of which are described), and that diets containing animal protein are generally more cholesterolemic and atherogenic (in rabbits, specifically) than diets containing vegetable protein. The protein effect can be modified by other dietary components (e.g., fiber, fat). Understanding the fatty acid (FA) metabolism of transition dairy cows is of importance due to the large amount of FA mobilized from adipose tissue, and high incidence of hyperketonemia (HYK), that occurs in situations when FA mobilization is excessive. In our first experiment, we observed differences in the plasma FA profile postpartum between HYK and nonHYK cows reflecting the increased mobilization of FA from adipose tissue in HYK cows There was a significant decrease in pyruvate carboxylase expression in cows that later developed HYK that resulted in a decreased pyruvate carboxylase to cytosolic phosphoenolpyruvate carboxykinase ratio. This may indicate a decrease in hepatic complete oxidative capacity of FA. There did not appear to be an effect of differences in FA profile between HYK and nonHYK cows on hepatic gene expression which was supported using primary hepatocytes in an in vitro model and changes in the FA profile of the media similar to that observed in transition dairy cows. Considering FA uptake by the mammary gland, there did not appear to be an effect of HYK, though there was a linear effect of arterial concentration on arterial-venous differences. In the final experiment, late lactation diets were fed that differed by 2 percentage units in dietary 16 versus 18 carbon FA. This change did not impact subcutaneous FA profile, nor the circulating plasma FA profile postpartum when FA are mobilized from adipose depots throughout the body. There were differences in adipose tissue FA profile between MP and PP cows, which could originate from difference in management, diet, number of mobilization and re-accretion cycles, a difference in metabolism, or another difference between parities. This dissertation provides new knowledge to the field of transition cow FA metabolism in relation to the liver, mammary gland, and adipose tissue. Future research in this area should focus on understanding the regulation of FA partitioning and metabolism to optimize hepatic and whole-body metabolic efficiency and health during the transition to lactation period.
