The aim of this book is to summarize our understanding on the insect nicotinic acetylcholine receptors. This area of research received great impetus from the identification of the first subunit sequences to be used as neonicotinoid insecticide target sites. Although a book of this nature can provide the details only of commonly published results, it is hoped that it may provide a useful guide to the newcomer to the field as well as to point out some of the future challenges. Abstract: The nicotinic acetylcholine receptor (nAChR) is a member of the ligand-gated ion channel superfamily of receptors. Each nAChR is composed of five subunits that assemble in combinations around a central channel to give the nAChRs many diverse properties. These receptors are classified into two groups, based on their location at the neuromuscular junction (NMJ) or in the nervous system. The primary function of neuronal nAChRs is to modulate synaptic transmission. Recent findings indicate that neuronal nAChRs play a role in the development of the nervous system. Because the nAChR has been implicated in nervous system development, the zebrafish (Danio rerio) has shown incredible potential as a model for the examination of the nAChR in these events. This manuscript is dedicated to describing the initial studies of the neuronal nAChR in the zebrafish model. Using RT-PCR and 5 & rsquo; and 3 & rsquo; RACE, our laboratory has cloned and sequenced the first neuronal nAChR subunit genes. To date, these include the & alpha;2, & beta;3, & alpha;7 and & alpha;8 nAChR subunits. Cloning of potential zebrafish nAChR & alpha;3 and & beta;4 subunit cDNAs is also currently underway. Using RT-PCR, the time course of expression of the zebrafish nAChR & alpha;2, & beta;3 and & alpha;7 RNAs has been determined. Each of these subunits was also localized using in situ hybridization with zebrafish embryos at specific time points. Ligand binding studies using the nAChR agonist epibatidine demonstrated the presence of assembled receptor subtypes by 2 and 5 days post fertilization (dpf), and expression of the zebrafish nAChR & alpha;7 subunit RNA in Xenopus oocytes provided evidence that these receptors Finally, apoptosis assays have been performed using nicotine and other cholinergic agents during different developmental time points to provide preliminary evidence of the effect nicotine may have on nervous system development. The work performed by our laboratory has provided the initial studies of the zebrafish neuronal nAChR, and provides further evidence for the use of this model in the study of the nAChR in development.

In the past few years, the scientific community has witnessed rapid and significant progress in the study of ion channels. Technological advance ment in biophysics, molecular biology, and immunology has been greatly accelerated, making it possible to conduct experiments that were deemed very difficult if not impossible in the past. For example, patch-clamp tech niques can now be used to measure ionic currents generated by almost any type of cell, thereby allowing us to analyze single-channel events. It is now possible to incorporate purified ion channel components into lipid bilayers to reconstitute an "excitable membrane." Gene cloning and monoclonal antibody techniques provide us with new approaches to the study of the molecular structure of ion channels. A variety of drugs have now been found or are suspected to interact with ion channels to exert therapeutic effects. In addition to the classical exam ples, as represented by local anesthetics, many other drugs, including cal cium antagonists, psychoactive drugs, cardiac drugs, and anticonvulsants, have been shown to alter the ion channel function. For certain pesticides such as pyrethroids and DDT, sodium channels are clearly the major target site. Many diseases of excitable tissues are known to be associated with, if not caused by, dysfunction of ion channels; these include cardiac ar rhythmias, angina pectoris, cystic fibrosis, myotonia, and epilepsies, to men tion only a few. Channel dysfunction can now be studied due to theoretical and technological developments in this area.

As part of its scientific activities, the German Research Council on Smoking and Health regularly provides opportunities for scientists to discuss progress in the field of nicotine research. In this context, the Research Council sponsored a Satellite Symposium in Hamburg, June 28-30, 1990 entitled "Effects of Nicotine on Biological Systems". This meeting was held in conjunction with the XIth International Congress of Pharmacology in Amsterdam and follows the first Satellite Symposium on Nicotine which was convened in Brisbane, Australia in 1987. The aim of these conferences has been to discuss state of the art research on the pharmacology and toxicology of nicotine and its metabolites and to integrate this information to help define nicotinic actions on the central and peripheral nervous system as well as to evaluate health or behavioral effects associated with use of this alkaloid. Furthermore, at this conference, potential therapeutic benefits of nicotine for certain disease states were discussed. Smoking and the health effects of smoking were dealt with only as far as they could not be separated from the effects of nicotine. This volume contains the lectures presented at the symposium and illustrates that knowledge of nicotine has advanced considerably in recent years with regard to mechanisms of its actions. Despite such progress however, it is apparent that a' large number of questions remain unanswered, especially in the light of new insight into cellular and molecular mechanisms which can be affected by nicotine.
