Mantle cell lymphoma (MCL) is an incurable, rare type of B-cell non-Hodgkin lymphoma, accounting for 5-6% of all lymphoma cases. A hallmark of MCL is the t(11;14)(q13;q32) translocation, juxtaposing the cyclin D 1 proto-oncogene (CCND1) to the immunoglobulin heavy chain gene (IgH), leading to CCND1 constitutive expression at the pre-B cell stage. Additionally, constitutively activated B cell receptor signaling (BCR) is required for the survival of MCL cells as demonstrated by the clinical use of selective inhibitors of BCR such as ibrutinib or acalabrutinib. De novo DNA methyltransferase 3A (DNMT3A), whose canonical function is to catalyze the transfer of methyl group onto the cytosine of unmethylated DNA, is known to be essential for the activation of B cells against pathogens and is highly expressed in a subset of MCL patients who are resistant to ibrutinib, suggesting that DNMT3A is a potential oncogene in MCL. In contrast, DNMT3A is reported as a tumor suppressor in acute myeloid leukemia (AML). In addition to the canonical function of DNA methylation, DNMT3A interacts with other epigenetic enzymes and transcription factors such as HDAC1 and PU.1 to regulate gene expression. However, little is known about the oncogenic functions of DNMT3A and its binding partners to regulate gene expression in the context of B-cell lymphoma. This thesis will examine the role of DNMT3A and propose the mechanisms by which it regulates gene expression in MCL. Additionally, I will test whether inhibiting DNMT3A by decitabine, an FDA-approved small molecule that leads to the degradation of all DNA methyltransferases (DNMTs), is a potential effective strategy in treating MCL. In the first chapter, I will discuss the current understanding of mantle cell lymphoma from both basic and translational perspectives. In chapter II, I will discuss the roles of DNMTs in normal B cell development and in hematological malignancies. In the third chapter, I will describe the experiments, the results, and my interpretations of the data to test the hypothesis that DNMT3A is required for the optimal growth of MCL cells. Here, I showed that DNMT3A is required for the optimal growth of MCL cells by regulating mitochondrial biogenesis independently of its methyltransferase activity. Mechanistically, DNMT3A induces the expression of MYC target genes through interaction with the transcription factors myocyte enhancer binding factor 2B (MEF2B) and MYC to enhance mitochondrial biogenesis and thus oxidative phosphorylation (OXPHOS). I further revealed that DNMT3A is involved in ibrutinib resistance as its expression is upregulated upon ibrutinib treatment in ibrutinib-resistant MCL cells and that the depletion of DNMT3A sensitizes these cells to ibrutinib. Targeting DNMT3A with low dose decitabine inhibits the growth of ibrutinib-resistant lymphoma cells and enhances the efficacy of ibrutinib both in vitro and in a patient-derived xenograft mouse model. In the last chapter, I will discuss the implications of my findings as well as unanswered questions for future studies. Most importantly, I will present evidence that DNMT3B is also a potential oncogene in mantle cell lymphoma and should be further studied.
