Kota Kolose (nama sebenarnya: Kolossai) berdekatan dengan Laodekia Hireapolis, yang terletak di pedalaman Asia. Kolose adalah satu kota di provinsi Romawi wilayah Asia. di bagian barat dari yang sekarang disebut Turki bagian Asia. Rekan sekerjanya, Epafras, yang melayani jemaat ini, mengunjungi sang rasul dan melaporkan perkembangan orang-orang percaya di sana dan munculnya sebuah ajaran sesat yang sedang merongrong mereka. Hal ini menunjukkan bahwa walaupun bukan Rasul Paulus sang pendirinya, tetapi tetap di bawah tanggung jawabnya. Ada tiga tokoh atau pemimpin yang disebutkan di kota Kolose ini, yaitu: Epafras, pendiri yang bijaksana (1:7; 4:12), Arkhipus, sahabat yang baik (4:17), dan Filemon, seorang Kristen yang terkemuka (Flm. 5-7). Kemungkinan Epafras adalah dari hasil penginjilan Rasul Paulus di Efesus yang menjadi rekan sepelayanan Paulus. Epafras inilah yang membawa kabar kepada Paulus tentang keberadaan jemaat di Kolose.
