Epilepsy is a disease characterized by spontaneous recurrent seizures. Underlying epilepsy is a state of neural hyperexcitability, which may be due to genetic predisposition or acquired after injury (e.g., infection, trauma). Following initial seizures, the regions of the brain involved undergo several anatomical and physiological changes that are collectively termed epileptogenesis. Among people with epilepsy, poor sleep is a common symptom. Reciprocally, it has been reported that sleep loss is among the most potent triggers for epileptic seizures. So, it appears that sleep loss and seizures participate in a vicious cycle in epilepsy. As the mechanisms of epileptogenesis and disease progression are continually elucidated, sleep dysregulation is emerging as a potential nexus of control over some of the broader pathologies associated with epilepsy. Conventional therapies are often ineffective in controlling seizures, especially in temporal lobe epilepsy (TLE). In TLE, a common site of seizure generation is the hippocampus. This thesis work attempts to disentangle the vicious cycle between sleep and seizures in TLE by independently investigating 1) the effects of 4-hour sleep deprivation (4SD) on GABAergic tonic and phasic inhibition in the hippocampus of seizure-naive mice, 2) the sleep responses to 4SD and hippocampal function in early epileptogenesis, and 3) sleep and circadian rhythms in a genetic model of TLE. This thesis work demonstrates that circadian sleep regulation, the response to 4SD, as well as tonic inhibition in the hippocampus are impaired at several points of epileptogenesis. Additional reduction of hippocampal tonic inhibition caused by transient sleep loss is a possible cause for the 4SD-induced increase in seizure susceptibility.
