For my dissertation work, I have focused on dissecting the mechanism and regulation of mRNA selection and recruitment to the human ribosome. I was interested in human eIF4B, which is a heavily phosphorylated, 69 kDa protein that promotes the mRNA helicase activity of eIF4A and interacts with the 43S pre-initiation complex. In Chapter 1, I introduce the mechanism by which translation initiation is regulated, focusing on control by post-translation modifications during growth and cellular stress. In Chapter 2, I describe a quantitative mass spectrometry method to characterize the site-specific phosphorylation changes that occur on eIF4G and eIF4B in response to mTOR inhibition in HeLa R19 cells. I also describe the development of a system to express and purify affinity tagged proteins from HeLa R19 stable cell lines. In addition to characterizing eIF4B phosphorylation dynamics, in Chapter 3 I characterize a novel 40S ribosomal subunit binding domain in eIF4B. I then described a cell-based assay I used to link this 40S binding domain to eIF4B's ability to associate with an mRNA. Described in Chapter 4, this method utilizes live cell fluorescence microscopy to track the ability of eIF4E, eIF4G, and eIF4B to accumulate into stress granules during oxidative stress. In Chapter 5, I characterize the conformation of eIF4B using ion mobility mass spectrometry and show that phosphorylation at Ser422 does not change its structure. In the final chapter, I outline the methods used to generate Flp-in T-REx HeLa R19 cell lines to stably express modified eIF4G and eIF4B constructs that are used in various experiments performed in this dissertation. My dissertation work builds upon the advances made to characterize phosphorylation sites on eIF4B to provide important mechanistic insight into the additional layer of regulation phosphorylation provides to control protein synthesis.
