Multivalent interactions are most frequently considered from an entropic perspective, attributed to entropically-assisted chelate type bonding. In order to obtain true thermodynamic parameters describing multivalency, a system must be in equilibrium and not kinetically-trapped in a non-equilibrium state. Divalent barium binding to EDTA and the conceptual fragments of EDTA serve as a powerful model of multivalency. Analysis of calorimetric data after the Jencks formalism reveals a favorable increment in interaction enthalpy of the binding for each acetate epitope; this behavior is attributed to relief of unfavorable coulombic repulsion in the unbound state. Comparison of the binding of chelates to Ca(II) and Ba(II) shows similar trends, although, the pentavalent DTPA lacks an increase in interaction energy when bound to Ca(II) due to an octahedral coordination geometry. Additionally, EDTA binds to Ba(II) with a lower affinity than Ca(II). This diminished affinity arises because the main structural elements in the Ba(II)-EDTA complex deviate significantly from the optimized geometries seen in the Ca(II)-EDTA complex.
