This study aims to compare traditional swabbing methods for collecting biological evidence from skin against a new instrument known as the M-Vac. Sexual assaults most often result in physical contact between the offender and the victim. In many cases, circumstances of the assault result in the victim not knowing where contact may have been made between themselves and the offender, leaving forensic practitioners unsure of where to sample. The M-Vac aims to eliminate the need for blind sampling by allowing practitioners to sample much larger surface areas than traditional swabbing methods permit. In this study, the traditional swabbing was compared against two M-Vac methods- a "localized" M-Vac method and a "whole arm" M-Vac method. Male saliva was placed on female skin and allowed to dry before collection using one of the three methods. Samples were then extracted, quantitated, and typed using STR analysis. The local M-Vac method visibly collected more male DNA than both other methods, though it was only statistically significant when 50uL of male saliva was originally placed on the females' skin. All methods resulted in a full male STR profile, which shows that the M-Vac is a viable option when blind swabbing would otherwise be utilized, therefore allowing for a larger surface area to be examined. This study briefly looked at the M-Vac's ability to collect touch DNA from female skin using a simulated strangulation model. Three "strangulation" time increments of 30, 60, and 90 seconds were used. Of these samples, 4 of the 12 trials resulted in a full male STR profile, while 6 of the remaining 8 resulted in a partial male profile. The last 2 samples resulted in no male STR presence. These results provide a basis for further investigation into the M-Vac's ability to collect touch DNA from skin compared to traditional collection methods.
