In Arabidopsis, the developmental transition from vegetative to reproductive growth is controlled by several genetic pathways. The autonomous pathway and the vernalization pathway converge at the regulation of FLOWERING LOCUS C (FLC) expression. Both activation and repression of FLC expression are controlled by epigenetic mechanisms. The first part of my thesis focuses on the study of loss-of-function mutants of SDG8 and SDG26, which encode SET domain proteins showing high homologies. Loss-of-function of SDG8, mutants sdg8-1 and sdg8-2, causes early flowering which is correlated with decreased expression of FLC and its homologues. By Western blot and ChIP, I demonstrated that levels of H3K36 di- and tri-methylation were greatly reduced while the level of H3K36 mono-methylation was increased in the sdg8 mutants. My study thus establishes that H3K36 di- and tri- but not mono-methylation is associated with transcription activation. In contrast to sdg8-1 and sdg8-2, sdg26-1 and sdg26-2 mutants showed a late flowering phenotype and an increased level of FLC expression. Nevertheless, molecular mechanism involved in FLC activation in sdg26-1 and sdg26-2 is currently unknown. In yeast, histone H2B mono-ubiquitylation is reported to precede H3K4 and H3K36 methylation during transcription initiation and elongation. The second part of my thesis focuses on molecular and functional characterization of AtUBC1 and AtUBC2 which encode homologues of Rad6, an E2 enzyme responsible for H2B mono-ubiquitylation in yeast. Although single mutants Atubc1-1 and Atubc2-1 have a normal phenotype, the double mutant Atubc1-1Atubc2-1 shows an early flowering phenotype. Our studies show that as in sdg8 mutants, expression of FLC and its homologues is reduced in Atubc1-1Atubc2-1. By Western blot, we showed that H2B mono-ubiquitination is significantly reduced in Atubc1-1Atubc2-1. Together these results suggest that H2B mono-ubiquitination and H3K36 methylation form a molecular pathway to regulate the FLC transcription, and play a crucial role in flowering. The last part of my thesis focuses on characterization of two novel RING domain genes LOC1 and LOC1, potentially involved in histone ubiquitination. I show that the double mutant loc1-1loc2-1 has a drastic phenotype of ectopic meristem formation. Molecular analysis revealed that the suppression of the Class I KNOX genes, STM, KNAT1/BP, KNAT2 and KNAT6, is released in loc1-1loc2-1. Physical and genetic interactions were observed between LOC1/LOC2 and CLF or LHP1, suggesting a Polycomb silencing pathway involved in repression of KNOX genes to confine shoot meristem.

The onset of flowering is an important step during the lifetime of a flowering plant. During the past two decades, there has been enormous progress in our understanding of how internal and external (environmental) cues control the transition to reproductive growth in plants. Many flowering time regulators have been identified from the model plant Arabidopsis thaliana. Most of them are assembled in regulatory pathways, which converge to central integrators which trigger the transition of the vegetative into an inflorescence meristem. For crop cultivation, the time of flowering is of upmost importance, because it determines yield. Phenotypic variation for this trait is largely controlled by genes, which were often modified during domestication or crop improvement. Understanding the genetic basis of flowering time regulation offers new opportunities for selection in plant breeding and for genome editing and genetic modification of crop species.
