This second book of the three-volume collection "Ion Transport in Tumor Biology" helps readers gain comprehensive knowledge of the pathophysiology of cancer. The authors highlight that ion transport proteins, channels and transporters - collectively referred to as the transportome - are significantly involved in the development and progression of cancer. Nearly 90% of malignant tumor diseases originate from epithelial cells, the function of which, for the most part, is based on the transportome. This volume focuses on molecular principles by showing that dysregulated expression and/or function of ion transporters have been correlated with malignancy in the vast majority of tumor diseases. The position of "Cancer Teaching Coordinator" at The University of Chicago has been a consistently rewarding one because of the enthusiasm and support of the faculty and the students. This volume is the result of the second of two recent intensive teach ing sessions which have been planned and implemented by the group which forms the Cancer Coordinator's Advisory Committee. The first of these teaching sessions was held in early March of 1964 and was entitled "LEUKEMIA, A Current and Forward Look. " It attracted overflow attendance from the students and staff of this medical institution augmented by members of the other medical centers in Chicago. It was a stimulating and instructive colloquium, and the only regret we heard expressed afterward was that we had not arranged for publication of the many excellent presentations. One of the events commemorating the seventy-fifth anniversary of the founding of The University of Chicago was this symposium on malignant transformations. This time the Committee advised us to plan on speedy publication, and, logically, it chose Dr. Werner Kirsten, a member of our faculty and an active and effective investigator in this general field of endeavor, to serve as editor of the volume. Again, two of the same ingredients for effective instruction were blended: an excellent group of scientists presenting their latest work and a fine, attentive audience of students and staff.

Cancer is a complex disease. Only 5-10% of human cancers are hereditary in nature. Many of us think of environmental agents when we think of carcinogens. The environment includes all that surrounds us, and environmental influences include not only chemical, physical and biological toxicants, but also diet and lifestyle. In this broadest sense, the environment contributes substantially in the development of human cancer. This book will describe how environment contributes to malignant transformation leading to profound changes in the genetic and signaling networks that control the functioning of the cell. It will critically discuss the understanding of the effects of environment on the development, progression and metastasis of cancer with current knowledge of the signaling networks that support functioning of transformed human cells. Genes and environmental factors that influence the origins of cancer are not necessarily the same as those that contribute to its progression and metastasis. Susceptibility gene variants for each specific cancer are being identified with emerging evidence of gene–environment interaction. Gene-environment interactions will be discussed through each specific cancer-based approach to address the question of how genetic variations can influence susceptibility to the individual type of cancer. It will also highlight and summarize epigenetic changes that increase the risk for susceptibility to a particular type of cancer, particularly in the presence of specific environmental factors. Thus, this book will contain chapters from the world’s experts focused on the current evidences that support the role of environment in the cancer etiology and in the growth of malignant lesions, and discuss who may be susceptible to environmental influences.
