Ab S. 48 beleuchtet Alison Abbott, wieso experimentelle Alzheimermedikamente in Studien bisher schlechter als erhofft abschnitten. Die Ursache liegt womöglich im Timing. Denn sobald ernste Schäden im Hirn entstanden sind, ist es zu spät, um ihre Folgen abzubremsen. Weitere Untersuchungen sollen nun zeigen, ob die Arzneien wirksamer sind, wenn Menschen sie vor dem Auftreten von Symptomen einnehmen. Die Ergebnisse werden in Wissenschaftskreisen mit Spannung erwar¬tet. Sollte die frühe Einnahme nicht die erwünschte Wirkung erzielen, steht nämlich nicht nur die Zukunft der Wirkstoffe auf dem Spiel, sondern auch die der führenden Theorie zur Ursache von Alzheimer, der Amyloid-Hypothese. Eine bestimmte Patientengruppe ist von vornherein von vielen dieser Medikamententests ausgeschlossen – entgegen der Verpflich¬tung zur Gleichbehandlung, die aus der UN-Behindertenrechtskonvention hervorgeht: Menschen mit Down-Syndrom. Warum Betroffene besonders häufig und früh Alzheimer entwickeln, erklärt unser Autor Anton Benz auf S. 54. Auch das gesunde Gehirn hält noch zahlreiche Rätsel für Neurowissenschaftler bereit. György Buzsáki vertritt ab S. 12 eine neue These, die so manche verbreitete Annahme der Hirnforschung über Bord wirft. Unser Geist startet demnach nicht als unbeschriebenes Blatt, sondern ist vielmehr voller anfangs sinnloser Einträge, die dann mit Eindrücken aus der Außenwelt verknüpft werden. Vor einigen Jahren ging ich zu nächtlicher Stunde allein durch die Stadt nach Hause. Es ist nicht weit und die frische Luft tut gut, dachte ich mir. Das bereute ich allerdings, als ich plötzlich hinter mir Schritte hörte, die immer schneller auf mich zukamen. Ich drehte mich um und sah im Dunkeln einen Mann, der augenscheinlich die Verfolgung aufnahm. Mein Herz raste, ich zitterte und beschleunigte. Mir schossen allerhand Gedanken durch den Kopf: Muss ich mich verteidigen? Wohin kann ich fliehen? Zum Glück fand in einem der Häuser dort eine Party statt; ich klingelte und man ließ mich ein. Wer weiß, vielleicht hatte er es ja nur eilig. Aber ich bin froh, dass ich es nicht darauf ankommen lassen musste, seine wahren Absichten herauszufinden. Für solche bedrohlich wirkenden Situationen hat uns die Evolution mit einem überlebenswichtigen Alarmsystem ausgestattet: Der Puls rast, Adrenalin flutet den Körper, wir reagieren automatisiert mit Flucht, Erstarren oder Verteidigung – begleitet vom intensiven Gefühl der Furcht. Jahrzehntelang galt der Mandelkern, wissenschaftlich Amygdala, als das Hirnzentrum für Angst und Schrecken, als Ort, wo solche Empfindungen entstehen. Studien an Tieren und Menschen schienen das zu belegen. Aber selbst Joseph LeDoux, jener Forscher, der die Idee einst befeuerte, bemüht sich heute darum, diese Darstellung zu korrigieren. So erzeugt der Mandelkern zwar die körperliche Reaktion auf Bedrohung, aber für das Gefühl der Angst, das Bewusstwerden der Gefahr ist er nicht zuständig. Auch sah man in einer defekten Amygdala lange die Ursache für Psychopathie. Doch kann es so einfach sein, wo das Gehirn derart komplex ist, fragt unser Autor Jan Schwenkenbecher und geht ab S. 68 dem »Mythos Amygdala« nach. Der Fall des vermeintlichen Angstzentrums zeigt zudem: Mitunter gibt es in der Forschung die Tendenz, gewünschte oder erwartete Effekte zu berichten und fehlende Befunde zu ignorieren. Ich habe eine Erinnerung an einen Ort, an den man eigentlich keine haben sollte. Sie ist nur sehr vage und geht ungefähr so: Durch eine verschwommene Fensterfront fällt Licht, ich höre ein Piepsen. Jemand ruft: »Sie wacht auf!« Es folgt hektisches Treiben, und dann ist die Szene auch schon wieder vorbei. Vor vielen Jahren bin ich im Operationssaal kurz aus der Narkose aufgewacht. Ich hatte weder Schmerzen noch habe ich Blut gesehen - insofern ist der Gedanke durchaus erträglich. In der Rückschau bietet er sogar einen interessanten Blick auf die Natur unseres Bewusstseins: Man kann es gezielt aus- und einschalten, und zwar unabhängig vom Schmerzempfinden. Außerdem gibt es eine ganze Bandbreite von Ausprägungen: von der Minimalversion, dem so genannten »minimal conscious state« (MCS), über Schläfrigkeit bis hin zur hoch konzentrierten Wachheit. Wie genau subjektives Erleben im Gehirn entsteht, ist allerdings nach wie vor eines der größten Rätsel der Wissenschaft. In diesem Dossier tauchen wir daher ab in die Welt des Bewusstseins und erkunden, was die Forschung hierzu bislang herausgefunden hat. An argument for a Copernican revolution in our consideration of mental features—a shift in which the world-brain problem supersedes the mind-body problem. Philosophers have long debated the mind-body problem—whether to attribute such mental features as consciousness to mind or to body. Meanwhile, neuroscientists search for empirical answers, seeking neural correlates for consciousness, self, and free will. In this book, Georg Northoff does not propose new solutions to the mind-body problem; instead, he questions the problem itself, arguing that it is an empirically, ontologically, and conceptually implausible way to address the existence and reality of mental features. We are better off, he contends, by addressing consciousness and other mental features in terms of the relationship between world and brain; philosophers should consider the world-brain problem rather than the mind-body problem. This calls for a Copernican shift in vantage point—from within the mind or brain to beyond the brain—in our consideration of mental features. Northoff, a neuroscientist, psychiatrist, and philosopher, explains that empirical evidence suggests that the brain's spontaneous activity and its spatiotemporal structure are central to aligning and integrating the brain within the world. This spatiotemporal structure allows the brain to extend beyond itself into body and world, creating the “world-brain relation” that is central to mental features. Northoff makes his argument in empirical, ontological, and epistemic-methodological terms. He discusses current models of the brain and applies these models to recent data on neuronal features underlying consciousness and proposes the world-brain relation as the ontological predisposition for consciousness.
