The application of host plant root leachates to potato cyst nematode- (PCN) infested soil in the absence of potato plants in the field gave approximately a 50% reduction in PCN population size. This control was due to a combination of hatch and in-egg mortality, the latter caused by apparently incomplete hatch stimulation of certain eggs, which were unable to respond fully (to eclosion) upon stimulation by root leachates. Laboratory experiments with soil columns demonstrated that the hatching factors in host plant root leachate were extremely mobile in soil, the most mobile hatching factors being G. pallida- selective. The degree of selectivity of the most mobile hatching factors towards G. pallida compared to G. rostochiensis was found to increase with increasing soil organic matter content. Ten hatching factors were isolated from potato root leachate, all with a common molecular weight of 530.5. HF E was found to be hatch active in vitro at concentrations as low as 2.1 x 10-8 M. The hatching activity of potato root leachates from segregants of crosses between potato and wild solanum species towards G. rostochiensis and G. pallida was found to segregate independently. Hatching factor fractionation studies with S. tuberosum x S. sucrense hybrid clones demonstrated segregation and recombination of hatching factors from the parents. The greater the proportion of tuberosum- derived hatching factors, the higher the hatching activity towards G. rostochiensis. The annual decline rate in the first and second year in rotation of G. rostochiensis in Co. Cork was calculated at 57% and 40%, respectively. Spontaneous hatch accounted for between 75 - 80% of this decline, with the remainder being due to in-egg mortality, presumably as a result of biological control. The number of culturable bacteria and the composition of the bacterial microflora in the cysts was found to increase with increasing soil temperature; no association was observed between the proportion of microflora expressing a particular lytic enzyme activity, and in-egg mortality.

When Franklin published her book on cyst nematodes in 1951, the cyst nematodes were already known as serious pests of brassicas, cereals, potatoes and sugar beets. However, at that time this group of nematode, with about 12 species, was considered tobe largely temperate in distribu tion. Now a total of 105 species (including those that are considered as synonyms or species inquirende by some or all) within six genera of cyst nematodes have been described from temperate, tropical, and subtropical regions and at least five species are important constraints to crop produc tion in tropical agriculture. The previous impression of localization of cyst nematodes in the temperate region was seemingly an artifact due to a greater concentration of nematologists in the temperate regions. Based on my own experience of working in several Asian and African countries, I believe that many more undescribed species are present in the tropical countries of Asia and Africa, and probably in other tropical regions. Most growers, extension workers, and research managers in these regions are still not aware of the possible harm of presence of these nematodes in their agricultural soils. The cyst nematodes are perhaps smaller than the smallest available computer chip but they are very well programmed to survive and pro pagate despite severe hardships. These nematodes are very selective in their choice of food; about 50% of known species are parasites of plants mainly in the families Poaceae and Fabaceae.

The hatching mechanisms of the two PCN species (G. pallida and G. rostochiensis) were compared and contrasted. The hatches of both PCN species to the root leachates from the hosts potato (PRL) and the tomato (TRL) and to artificial and natural HFs produced dose-response profiles that exhibited multiple peaks of hatch, termed the oligomodal hatch response. In most cases a high-concentration (main peak) activity was combined with one or more low-concentration ancillary peaks of hatch, active down to HF concentrations as low as 10-15 g 1-1. The ancillary peak was observed only with encysted, not mechanically freed eggs, and possible influenced by communication between hatched and unhatched J2s. In the bioassay, the ancillary peaks occurred after two weeks incubation, compared to the main peak, which occurred after one week. Expression of the ancillary peak was influenced seasonally by both the conditions of the unhatched eggs and leachate. The ancillary peak response was exhibited by eggs starting in May, corresponding to the beginning of the traditional cultivation period of the potato. The oligomodal dose-response phenomenon was restricted to leachates from plants aged eleven days or older. G. pallida hatched more freely that G. rostochiensis in PRL from up to ten-day old plants, from which point G. rostochiensis dominated. Studies performed with individual natural and artificial PCN HFs showed that single HF preparations could induce an oligomodal hatch response. In general, G. rostochiensis responded to more different HFs than did G. pallida. Studies with the GCA (glycoalkaloid) HFs demonstrated a correlation between reported GCA sterol-binding affinity and G. rostochiensis hatching activity. Other non-GCA HFs attributed with sterol-specific activity, the polyene antibiotics, induced high levels of hatch of both G. pallida and G. rostochiensis. Polyene-induced hatch was found to be dependent on intact polyene structure, an essential feature for reported sterol-mediated membrane perturbation. A model GCA hatching mechanism was offered: the GCA HFs initiate hatch after complexing with an eggshell membrane sterol, possibly leading to the formation of a pore, which may facilitate the efflux of Ca2+, ultimately resulting in hatch. GCA HFs a-solanine and a-chaconine were detected at hatch inducing concentrations in PRL by ELISA. Association studies between artificial and natural HFs and PRL revealed that PCN HFs can exhibit HS (synergy) and HI (inhibition) activity, in addition to HF activity (in the absence of PRL), depending on concentration, attributable to partial agonistic activity.

In 1992 a Concerted Action Programme (CAP) was initiated by Peter Sijmons with the purpose of intensifying collaborations between 16 European laboratories working on plant-parasitic nematodes. The four-year programme entitled Resistance Mechanisms Against Plant-Parasitic Nematodes focused on molecular aspects of the interaction between sedentary nematodes and plants on the model system Arabidopsis and on novel resistance strategies. Funding was provided mainly for exchange visits between collaborating laboratories and for the organization of annual meetings. During the last annual meeting which was held in May 1996 in Toledo, Spain, Carmen Fenoll initiated the production of this volume.
