The microcirculation plays varied and critical roles in our general well being and defects in the microcirculation manifest themselves in many important disease states. In this thesis, the extension of Doppler ultrasound to the ultrasound frequency range 20-200 MHz is investigated as a non-invasive and quantitative technique for detecting, measuring, and imaging the microcirculation. The theoretical advantages and limitations of high-frequency Doppler ultrasound (HFD) are discussed. Experimental continuous-wave and pulsed-wave HFD systems, which have been designed, constructed and optimised for detecting blood flow in the microcirculation, are described. Implementation of the systems at 40-MHz and 50-MHz in experimental investigations with phantoms show that HFD can detect and measure velocities on the order of the blood velocities found in the capillaries (

Microbubble ultrasound contrast agents have shown clinical potential for characterising blood flow in the microcirculation using 1 to 10 MHz ultrasound; however, scaling their use for mouse imaging with high frequency ultrasound (10 to 60 MHz) has not been addressed. The goal was to determine the utility of microbubbles for contrast flow studies in the mouse with 30 MHz ultrasound by investigating their acoustic response in vitro and optimal dose in vivo. In vitro results showed that microbubble attenuation and backscatter increased linearly for concentrations between 2.4 x 106 and 2.8 x 107 bubbles per mL. In vivo results showed that the peak enhancement of the agent increased linearly for doses between 10 and 60 muL kg -1, the duration of enhancement varied between 200 to 300 seconds and the integrated enhancement (area under the curve) increased linearly up to 100 muL kg-1. These doses corresponded to the range of concentrations used in vitro. However, optimisation of the reproducibility of this technique still needs to be addressed.

Recent evidence suggests that radiation may have a significant effect on tumour vasculature in addition to damaging tumour cell DNA. It is well established that endothelial cells are among the first cells to respond after administration of ionizing radiation in both normal and tumour tissues. It has also been suggested that microvascular dysfunction may regulate tumour response to radiotherapy at high doses. However, due to limitations in imaging the microcirculation this response is not well characterized.Advances in high-frequency ultrasound and computation methods now make it possible to acquire and analyze 3-D ultrasound data of tumour blood flow in tumour microcirculation.This thesis outlines the work done to test the hypothesis that single dose 8 Gy radiotherapy produces changes in tumour blood vessels which can be quantified using high-frequency power Doppler ultrasound. In addition, the issue of reproducibility of power Doppler measurements and the relationship between histopathology and power Doppler measurements have been examined.
