Calorie restriction (CR) without malnutrition has been shown to increase lifespan across numerous animal models. CR is associated with a metabolic shift away from glycolysis and toward fatty acid beta-oxidation. Given that low carbohydrate (LC) diets induce similar metabolic changes as CR, restriction of dietary carbohydrate may induce many of the same favorable changes in health and longevity as CR. The objective of this study was to determine the influence of LC and ketogenic (KT) diets on longevity and healthspan in mice. To investigate this, C57BL/6 mice were maintained on a fixed energy intake (11.2 kcal/day) and fed a KT diet (90% calories from fat, 10% from protein), LC diet (70% calories from fat, 10% from protein), or control diet (CT, 17% calories from fat, 18% from protein). Mice were started on the diets at 12 months of age and either allowed to live their natural lifespan or tested for physiological function after 1 month or 14 months of dietary intervention.
