Plants are sessile, highly sensitive organisms that actively compete for environmental resources both above and below the ground. They assess their surroundings, estimate how much energy they need for particular goals, and then realise the optimum variant. They take measures to control certain environmental resources. They perceive themselves and can distinguish between ‘self’ and ‘non-self’. They process and evaluate information and then modify their behaviour accordingly. These highly diverse competences are made possible by parallel sign(alling)-mediated communication processes within the plant body (intraorganismic), between the same, related and different species (interorganismic), and between plants and non-plant organisms (transorganismic). Intraorganismic communication involves sign-mediated interactions within cells (intracellular) and between cells (intercellular). This is crucial in coordinating growth and development, shape and dynamics. Such communication must function both on the local level and between widely separated plant parts. This allows plants to coordinate appropriate response behaviours in a differentiated manner, depending on their current developmental status and physiological influences. Due to their sessile lifestyle, plants need to efficiently adapt to changing environmental conditions during their life cycle. Nutrient acquisition from the soil has to be able to adapt to considerable fluctuations in concentrations to ensure adequate distribution between tissues, cells and organelles. The storage and retrieval of nutrients, metabolites or toxic substances in vacuoles plays an important part in cellular homeostasis in plants. The long-range transport and maintenance of turgor is critically dependent on the availability of water and rate of evaporation, while at the same time photosynthetic products have to be transported to all plant parts. As a result plants contain a large number of ATP-dependent pumps and secondary transporters that, in order to adapt to the changing environment, need to be regulated by a complex network of sensing and signaling mechanisms. Plants share many basic elements of signal transduction with animals, but also contain plant-specific signaling molecules and mechanisms. In this volume, the role of transporters and pumps in the regulation of movement, long-range transport and compartmentalization of water, solutes, nutrients and classical signaling molecules is highlighted, and the function, regulation and membrane-transporter interaction and their roles in plant signaling controlling plant physiology and development are discussed.

Plants contend with a myriad of environmental stressors, including insect herbivory. To defend against insect herbivory, plants employ defense responses to mitigate damage. Insects are mobile organisms, therefore, wounded tissues in plants send systemic signals to engage defense mechanisms in unwounded tissues to attenuate further herbivory. These defense signals are mechanistically complex. Long-distance electrical signals are speculated to convey the wound signal to distal tissues. Ionotropic glutamate receptors have previously been identified as key components of the electrical defense signal, but additional molecular components are expected to play a role as well. To uncover new molecular players, I performed a genetic screen which revealed that the mechanosensitive ion channel, MSL10, is important for wound induced long distance signaling. In Chapter 1, I review wound signaling in plants and explore the various mechanisms proposed to drive long-distance signals. Chapter 2 describes the discovery of the MSL10 phenotype and characterization of its role in electrical wound signaling. In Chapter 3, I explore the cell biology of MSL10 and how localization in the vasculature is important for signaling. I employed genetically encoded biosensors to study signaling events in plants to better understand their spatial patterning in Chapter 4. Finally, Chapter 5 summarizes the key findings of the thesis project and proposes new avenues of research to answer new open questions. Taken together, this work within the thesis provides a new foundation of knowledge for long-distance plant signaling and opens exciting new areas of study.

Oxygen (O ) appeared in significant amounts in the Earth’s atmosphere over 2. 2 2 billion years ago, largely due to the evolution of photosynthesis by cyanobacteria (Halliwell 2006). The O molecule is a free radical, as it has two impaired electrons 2 that have the same spin quantum number. This spin restriction makes O prefer to 2 accept its electrons one at a time, leading to the generation of the so-called reactive oxygen species (ROS). The chemical nature of these species dictates that they can create damage in cells. This has contributed to the creation of the “oxidative stress” concept; in this view, ROS are unavoidable toxic products of O metabolism and 2 aerobic organisms have evolved antioxidant defences to protect against this tox- ity (Halliwell 1981; Fridovich 1998). Indeed, even in present-day plants, which are full of antioxidants, much of the protein synthetic activity of chloroplasts is used to replace oxidatively damaged D1 and other proteins (Halliwell 2006). Yet, the use of the “oxidative stress” term implies that ROS exert their effects through indiscriminate widespread inactivation of cellular functions. In this context, ROS must not be able to react with lipids, proteins or nucleic acids in order to avoid any damage to vital cellular components. However, genetic evidence has suggested that, in planta, purely physicoche- cal damage may be more limited than previously thought (Foyer and Noctor 2005).
