The first symposium on metabolic compartmentation in brain was held at the Rockefeller Foundation, Bellagio, Italy, July 11-16, 1971; the proceedings CR. Balazs and J. E. Cremer, editors, MacMillan) appeared in 1973. At the conclusion of the first symposium it was decided to assess in 2-3 years the progress in this rapidly developing area. This volume represents the proceedings of an Advanced Study Institute, made possible by a grant from the North Atlantic Treaty Organization, Scientific Affairs Division. Schneider xi Introduction The term metabolic compartmentation, as related to the chemistry of the brain, appeared in print for the first time about 15 years ago. The concept was developed in the laboratory of Dr. Heinrich Waelsch as a result of studies related to the metabolism of glutamic acid and glutamine. It was welcomed by a number of neurochemists who felt that the concept had validity and would help explain metabolic phenomena that were otherwise quite puzzling. The concept gradually achieved general acceptance, and by 1971 the amount of information that could profit by being examined from the point of view of metabolic compartmentation had increased sufficiently to warrant a symposium on the subject ("Metabolic Compartmentation in the Brain," Balazs and Cremer, eds. , MacMillan, 1973). Almost all the participants at that initial symposium were present at the second, on which this volume is based.

The NATO Advanced Study Institute held in the Hotel Metropole Llandrindod Wells, Powys, Wales from July 26th to August 4th 1981 provided an excellent platform for presentation and appraisal of our current knowledge of neurotransmitter systems. The proceed ings comprise the present volume and these were arranged to allow study of the pathways and the interactions of specific transmitter systems, one with another, in the various sub-regions of the central nervous system. In addition several of the key factors involved in neurotransmitter release were highlighted. These include the structural and molecular organisation of the nerve terminal, its organelles (such as vesicles, neurotubules and synaptic densities) and the mechanisms involved in neurotransmitter release including the participation of calcium. The organisation of transmitter synthesis, storage and transport were discussed together with their linkage to other streams of metabolism, and the cellular compartmentation of these processes. In addition to the principal lectures given at the Institute, many part1c1pants presented short reports for discussion and these are all included in this volume. The success of the Study Institute was due to the careful planning of the scientific programme to allow a smooth flow and integration of the topics included and this is reflected in the organisation of this book. This volume is concerned with metabolic reactions occurring in the nervous system. Some time ago, it was thought that since most of the intermediary metabolism that can be observed in the brain is not specific to this organ, there is little justification in studying neural metabolism as such. Later it was realized that for an understanding of neural functions, the understanding of metabolism in the brain and its alterations is essential. All aspects of the metabolism of a substrate in brain, or all metabolic reactions of the nervous system, could not be included in this volume; some will be dealt with in other volumes (such as the ones covering metabolic turn over, alterations of metabolism, or pathology). Review of the aspects covered here clearly shows that the study of metabolic reactions in the nervous system is a very active field, producing important results. As in so many areas of research, as we learn more, new aspects become known, new questions emerge, and we see that in solving some problems we open areas with many additional problems to solve. But the accomplishments to date are impressive and indicate further important advances in the future. Brain metabolism is more active, more plastic, and more comprehensive than previously estimated. It is an essential part of brain function, and with its alteration, brain function will be altered. This shows the importance of more knowledge in this area. It is hoped that this volume will be of assistance in such further studies.
