Many common human diseases have a multifactorial origin: they are influenced by a person's genetic predisposition as well as by factors in the environment. This volume deals with the application of recombinant DNA techniques to the identification of diseases that have more than one inherited component. Focus is on the polygenic factors responsible for coronary atherosclerosis. Several other disorders having a polygenic origin are also discussed, including hypertension, diabetes mellitus, psychiatric diseases, and autoimmune (HLA-related) disorders. Problems raised by the study of different families or different populations are covered, as well as the possibility of applying molecular techniques to disease prevention--for example, through gene therapy. Also, some of the ethical issues that relate to human gene mapping are briefly explored.

Enormous technical advances in the field of eukaryotic genetics have provided the opportunity to study human biology as intensively as previous studies of prokaryotic biology. However, despite spectacular progress in our understanding of the molecular basis of human genetic diseases, they usually remain difficult to cure, and often fatal in the pediatric age group. This volume is timely in addressing new genetic approaches to studying and treating intractable diseases with known and unknown etiology. Heritable human genome editing - making changes to the genetic material of eggs, sperm, or any cells that lead to their development, including the cells of early embryos, and establishing a pregnancy - raises not only scientific and medical considerations but also a host of ethical, moral, and societal issues. Human embryos whose genomes have been edited should not be used to create a pregnancy until it is established that precise genomic changes can be made reliably and without introducing undesired changes - criteria that have not yet been met, says Heritable Human Genome Editing. From an international commission of the U.S. National Academy of Medicine, U.S. National Academy of Sciences, and the U.K.'s Royal Society, the report considers potential benefits, harms, and uncertainties associated with genome editing technologies and defines a translational pathway from rigorous preclinical research to initial clinical uses, should a country decide to permit such uses. The report specifies stringent preclinical and clinical requirements for establishing safety and efficacy, and for undertaking long-term monitoring of outcomes. Extensive national and international dialogue is needed before any country decides whether to permit clinical use of this technology, according to the report, which identifies essential elements of national and international scientific governance and oversight.

The Neanderthals populated western Europe from nearly 250,000 to 30,000 years ago when they disappeared from the archaeological record. In turn, populations of anatomically modern humans, Homo sapiens, came to dominate the area. Seeking to understand the nature of this replacement, which has become a hotly debated issue, Paul Mellars brings together an unprecedented amount of information on the behavior of Neanderthals. His comprehensive overview ranges from the evidence of tool manufacture and related patterns of lithic technology, through the issues of subsistence and settlement patterns, to the more controversial evidence for social organization, cognition, and intelligence. Mellars argues that previous attempts to characterize Neanderthal behavior as either "modern" or "ape-like" are both overstatements.
