Molecular imprinting is one of the most efficient methods to fabricate functional polymer structures with pre-defined molecular recognition selectivity. Molecularly imprinted polymers (MIPs) have been used as antibody and enzyme mimics in a large number of applications. The outstanding stability and straightforward preparation make MIPs ideal substitutes for biologically derived molecular recognition materials, especially for development of affinity separation systems, chemical sensors and high selectivity catalysts. New MIP materials are being increasingly applied to solve challenging problems in environmental sciences, food safety control, biotechnology and medical diagnostics. The synthesis of catalytically active polymers by MIP techniques results in tailor-made synthetic polymers, which are capable of subsequent molecular recognition of particular molecules. Even though considerable efforts have been expanded in the past to produce effective enzyme mimetic MIPs, catalytic rates and specificities have still been significantly lower, when compared to natural enzymes. The main reasons have been the use of unsuitable templates (i.e. those lacking particular molecular geometrics or stereochemistries) and suboptimal orientation of the appropriate active sites in the polymer matrix. Since the catalytic activity of a MIP is mediated through the cavity sites created in the macromolecular entity of the MIP, the generation of these active sites should be achieved through a particular polymerisation chemistry and utilising specific templates. Since control over the molecular architecture of the cavity sites in the MIPs is the decisive factors for those polymers to exhibit efficient catalytic activity and selectivity, templates that feature the appropriate functional and geometrical specification have to be designed and synthesised. A significant approach for successful achievement of this therefore should use the catalytic mechanism of the enzymes as a guideline. It has been described that the catalytic rate of biological and chemical reactions by enzymes is achieved when enzymes lower the activation energy of the reaction. This is achieved due to the differential binding energy of the substrate in its transition state (TS) and its ground state (GS). In this case, the higher binding affinity of the enzyme to the TS of the substrate compared to its GS enables the activation energy of the reaction to be lowered, and hence enhances the rate of the reaction.

The development of artificial catalysts is an exciting, emerging new area of chemical science and technology. Knowledge of enzyme structure and function is being used to help develop catalytically active molecularly imprinted polymers. By using coenzymes in imprinted polymers it is possible to mimic the catalytic activity of enzyme active sites. Coenzymes as imprinting templates create binding sites within a polymer, and these sites can be designed to have enzyme-mimetic catalytic activity. This work describes the creation of imprinted polymers with recognition sites for flavins and thiamines. In Chapter IV, a Riboflavin derivative (3) was non-covalently imprinted as a model to study the effect of varying the imprinting and template removal conditions. The template 3 was shown to be redox active, although whether the redox changes took place in the recognition site was inconclusive. In Chapter V, a two-point binding non-covalent molecularly imprinted polymer for 2 was made. Testing the recognition of various coenzymes and coenzyme analogs (2-6) has explored and defined the binding characteristics of the coenzyme-imprinted polymer. Thiamine and several analogs were studied for their potential as templates for an imprinted polymer for the recognition of thiamine in Chapter VI. The imprinting system has potential to be developed further in order to realize catalytically active systems. This dissertation includes my previously published material.
