Excerpt from Motor Memories: A Saga of Whirling Gears [The automobile, the airplane, the radio and the motion picture, all creations of the last thirty or forty years, have revolutionized our American scene and way of life; yet Detroit - the home of the automobile - has done nothing to preserve the records of this monumental achievement and, unless timely measures are taken to secure them from oblivion, the knowledge of the development of the automobile and its inventions will perish with the pioneers, in whose memories, alone, it today exists ... Third, I examine how the rate of adaptation to changes in the environment is modulated by the statistics of that environment. Specifically, I show how the rate at which the motor system adapts to changes in the environment is primarily determined not by the degree to which environment change occurs - i.e. environmental variability - but by the degree to which the changes that do occur persist from one movement to the next - i.e. the trial-to-trial consistency of the environment. Finally, I find that repetition of the same learning stimulus can greatly potentiate the effect of consistency, although, unlike consistency, repetition alone does not increase adaptation rate.

Movement and memory are two of the most fundamental components of our existence. From the moment of birth, we rely on a variety of movements to interact with people and objects around us, and as we grow, we continuously form new motor memories to improve the fidelity of these interactions by exploring and learning more about our environment, especially in unfamiliar situations, ultimately becoming better equipped to handle novel and unknown environments.

"Background: Motor memory consolidation is a process by which motor memories encoded during the practice of a motor skill are transformed from an initial fragile state to a more solid and stable state. Motor memories encoded by older adults are easily disrupted when another task is practiced soon after motor practice, during the first stages of consolidation. However, the neural mechanisms underlying this higher susceptibility to memory interference in older individuals are not known. Transcranial magnetic stimulation (TMS) is a non-invasive brain stimulation (NIBS) technique that can be used to manipulate brain activity thus providing insights into the mechanisms involved in memory formation processes. Previous studies with young adults have shown that the primary motor cortex (M1) is essential for the first stages of motor memory consolidation. However, whether this area of the brain also hosts the neural substrates subserving the early phases of motor memory consolidation in older adults is not known. Continuous theta-burst (cTBS) applied with TMS suppresses brain excitability and can theoretically interfere with the consolidation of motor memory. Objective: To determine if chronological age increases the susceptibility to motor memory interference induced with TMS on M1 excitability. Hypotheses: Applying cTBS over M1, immediately after motor practice, will disrupt memory consolidation reducing off-line gains in skill level in both young and older groups when tested 7 hours after practice. However, the older group will show a larger decline compared to the young group, confirming that aging increases the susceptibility to motor memory interference and that M1 is involved also in the elderly. Furthermore, we hypothesize that in contrast to older individuals, young subjects receiving cTBS will be able to recover the motor memory through a night of sleep, showing improvements in skill level 24 hours after practice. Methods: Fifty healthy subjects (25 young and 25 older adults) were recruited. Immediately after practicing an implicit version of the serial reaction time task (SRTT), each age group was randomized into 2 sub-groups. Either a continuous theta-burst stimulation protocol (cTBS) or a sham cTBS protocol was applied with TMS on M1 immediately after practicing the SRTT. Retention tests of the SRTT were performed 7 and 24 hours after motor practice. Performed after a wakeful period (7 hours) and after a night of sleep (24 hours), these two retention tests allowed us to determine if aging affects sleep-dependent and independent memory consolidation processes to the same extent. Results and Conclusion: Results from the current study demonstrated that: (1) Young subjects were significantly better in skill learning compared to older subjects. (2) cTBS applied over M1 disrupted the consolidation of sequential memory in young subjects. (3) Young subjects that received cTBS could rescue the disrupted sequential memory over a night of sleep. (4) Older subjects appeared to be protected from the disruptive effects of cTBS on sequential memory.Relevance: M1 is an important therapeutic target area in individuals with neuromotor deficits such as patients with stroke or Parkinson's disease. Also, M1 has been identified as a critical area of the brain for the early stages of motor memory consolidation in younger individuals (Muellbacher et al. 2000). Considering motor memory consolidation impairment is parallel to decline in skill performance in older adults (Censor et al. 2011); Gaining insight into the role of this area of the brain in memory consolidation in older adults may improve our capacity to design more effective NIBS strategies to improve the acquisition of motor skills in these individuals.
