The book offers readers an understanding of the development of neural crest cells, which is crucial as many birth defects and tumours are of neural crest origin. The neural crest is a transient tissue of the vertebrate embryo. It originates from the future spinal cord and neural crest stem cells emigrate from this location to various places in the embryo, giving rise to many different cell types and tissues. Neural crest derivatives include the peripheral nervous systems, endocrine cells such as the adrenal medulla, smooth musculature of the cardiac outflow tract and great blood vessels, as well as craniofacial bone and cartilage. The underlying mechanisms that regulate embryonic neural crest development are still being investigated and are important for our understanding of neural crest pathologies. Readers will have ready access to current research topics, elaborated in great detail, with a focus on adult neural crest-derived stem cells, which persist in various locations of the postnatal organism. Delving into stem cells from different locations of the body, the book explores the best possible source of such cells for future use in medical applications.

Developing the second edition of Neural Development and Stem Cells was neces- tated by the rapid increase in our knowledge of the development of the nervous system. It has become increasingly clear that stem cells are a heterogeneous population that changes extensively during development. Perhaps the most important advance in our understanding of stem cell behavior has been the realization that regionalization of stem cells occurs early in development and this bias toward differentiation in phe- types of neurons or cells characteristic of a particular part of the brain appears to persist even after prolonged culture. We have therefore included additional chapters on olf- tory epithelial stem cells and retinal stem cells, both of which differ in their properties from ventricular zone and subventricular zone–derived neural stem cells. It is also now clear from an analysis of mutants and transgenics where the death or self-renewal pa- way is altered that cell death regulates stem cell number. As a consequence, this second edition includes a separate chapter on cell death that summarizes the important changes in the death pathway that occur as stem cells mature. The existing chapters in the book have also been extensively revised and updated by experts who have generously c- tributed their time and expertise. The chapters have been organized along the lines of our understanding of how the nervous system develops (Fig. 1, on p. vi).

This 1999 edition of The Neural Crest contains comprehensive information about the neural crest, a structure unique to the vertebrate embryo, which has only a transient existence in early embryonic life. The ontogeny of the neural crest embodies the most important issues in developmental biology, as the neural crest is considered to have played a crucial role in evolution of the vertebrate phylum. Data that analyse neural crest ontogeny in murine and zebrafish embryos have been included in this revision. This revised edition also takes advantage of recent advances in our understanding of markers of neural crest cell subpopulations, and a full chapter is now devoted to cell lineage analysis. The major research breakthrough since the first edition has been the introduction of molecular biology to neural crest research, enabling an elucidation of many molecular mechanisms of neural crest development. This book is essential reading for students and researchers in developmental biology, cell biology, and neuroscience.

The neural crest is a remarkable embryonic population of cells found only in vertebrates and has the potential to give rise to many different cell types contributing throughout the body. These derivatives range from the mesenchymal bone and cartilage comprising the facial skeleton, to neuronal derivatives of the peripheral sensory and autonomic nervous systems, to melanocytes throughout the body, and to smooth muscle of the great arteries of the heart. For these cells to correctly progress from an unspecifi ed, nonmigratory population to a wide array of dynamic, differentiated cell types—some of which retain stem cell characteristics presumably to replenish these derivatives—requires a complex network of molecular switches to control the gene programs giving these cells their defi ning structural, enzymatic, migratory, and signaling capacities. This review will bring together current knowledge of neural crest-specifi c transcription factors governing these progressions throughout the course of development. A more thorough understanding of the mechanisms of transcriptional control in differentiation will aid in strategies designed to push undifferentiated cells toward a particular lineage, and unraveling these processes will help toward reprogramming cells from a differentiated to a more naive state. Table of Contents: Introduction / AP Genes / bHLH Genes / ETS Genes / Fox Genes / Homeobox Genes / Hox Genes / Lim Genes / Pax Genes / POU Domain Genes / RAR/RXR Genes / Smad Genes / Sox Genes / Zinc Finger Genes / Other Miscellaneous Genes / References / Author Biographies

This expanded and updated edition reflects the enormous increase in our knowledge of the development of the nervous system and the use of neural stem cells in clinical applications. Integrating new information about the use of development of neural stem cells for clinical applications, Drs. Rao, Carpenter and Vemuri have added additional chapters on neural transplantation and development of neural stem cells using reprogramming technologies. A new chapter on cancer in nervous system delineates how the fundamental biology of neural stem cells informs our understanding of cancer and tumor formation in the nervous system. Special attention is paid to the derivation of neural cells from embryonic stem cells and induced pluripotent stem cells. Authoritative and up-to-date, Neural Development and Stem Cells, 3rd Edition provides neuroscientists with a handy guide to stem cells in the nervous system, tracing with great clarity the development of stem cells from differentiation to neurons, astrocytes, and oligodendrocytes.

Active neuroscientists survey NSCs as potential tools for central nervous system and spinal cord repair by explaining their clinically significant fundamental properties, manipulations, and potential therapeutic paradigms. Their discussion of the fundamental biology of NSCs illustrates the signaling pathways that regulate stem cell division and differentiation, and defines the methods of NSC expansion and propagation, neuromorphogenesis, the factors determining cell fate both in vitro and in situ, and the induction of self-reparative processes within the brain. They also present strategies that may lead to fruitful clinical applications in the near future. These range from the replacement of degenerated, dysfunctional, or maldeveloped cells to the provision of factors that may protect, correct, recruit, promote self-repair, or mediate the connectivity of host cells.
