Prior to 1979, consideration of the problem of the carcinogenicity of the aromatic amine class of chemicals took place primarily in poster sessions and symposia of annual meetings of the American Association for Cancer Research and analogous international associations. In November 1979 the first meeting concerned with the aromatic amines was held in Rockville, Haryland under primary sponsorship of the National Cancer Institute. The proceedings from this meeting were published as Monograph 58 of the Journal of the National Cancel' Institute in 1981. The second meeting in this series, the Second International Conference on N-Substituted Aryl Compounds, was held in March/April of 1982 in Hot Springs, Arkansas. The National Cancer Institute and The National Center for Toxicological Research were the primary sponsors of this meeting. The proceedings were published as Volume 49 of the journal En-vil'onmental Health Perspectives in 1983. The third meeting in this series was held in April of 1987 at the Dearborn Hyatt in Dearborn, Michigan. The principal sponsor of this meeting was the Heyer L. Pre ntis Comprehensive Cancer Center of Metropolitan Detroit. The proceedings, Carcinogenic and Mutagenic Responses to Aromatic Amines and Nitroal'enes, were published in 1987 by Elsevier Press. The fourth meeting was held in Cleveland, Ohio, on July 15-19, 1989.

Nitroaromatics are widely available synthetic building blocks which present a strategical opportunity to serve as direct precursors to nitrogen-containing molecules of increasing complexity and worth through reductive/deoxygenative methods. Trivalent phosphorus compounds are valuable stoichiometric reagents for a range of reductive O-atom transfer reactions involving the conversion of R3P[superscript III] to R3P[superscript V]=O (including nitroarene deoxygenation), but can suffer from instability, pyrophoricity, and difficulty of removal during purification for both the phosphine and the generated phosphine oxide. Having the ability to start with a bench-stable phosphine oxide--which most often is regarded as a waste by-product--and repeatedly generate an active phosphine species in situ for catalytic reaction chemistry is a motivating concept with potentially practical benefits. With the incorporation of a hydrosilane reductant, it is demonstrated that a small-ring cyclic phosphine oxide can be quickly reduced in situ to catalyze the intramolecular cyclization of o-functionalized nitrobenzene derivatives to produce nitrogen-containing heterocycles (2H-indazoles, 2H-benzotriazoles, carbazoles, indoles, and benzimidazoles), as well as the intermolecular C-N cross coupling of nitroarenes with boronic acids through exhaustive nitro deoxygenation via P[superscript III]/P[superscript V]=O catalysis. The work herein not only describes the discovery of new organocatalytic methods founded on the use of a designer, small-ring phosphine oxide (pre)catalyst (1,2,2,3,4,4-hexamethylphosphetane 1-oxide) for the reductive transformations of nitroarenes, but also details investigations into the reaction mechanism for both reductive cyclization and C-N coupling reactions.

The authors present evidence that the shock-initiation chemistry of nitroarenes is dominated by the intermolecular hydrogen transfer mechanism discussed previously. The acceleration by pressure, kinetic isotope effect, and product distribution are consistent with the bimolecular transition state kinetic isotope effect, and product distribution are consistent with the bimolecular transition state rather than rate-determining C-N homolysis. GC-MS analysis of samples which were subjected to a shock wave generated by detonation of nitromethane shows that nitrobenzene produces aniline and biphenyl, and o-nitrotoluene forms aniline, toluene, o-toluidine and o-cresol, but not anthranil, benzoxazinone, or cyanocyclopentandiene. In isotopic labeling experiments o-nitrotoluene and TNT show extensive H-D exchange on their methyl groups, and C-N bond rupture is not consistent with the formation of aniline from nitrobenzene or nitrotoluene, nor the formation of o-toluidine from o-nitrotoluene. Recent work incorporating fast TOF mass spectroscopy of samples shocked and quenched by adiabatic expansion shows that the initial chemical reactions in shocked solid nitroaromatic explosives proceed along this path.
