Negative strand RNA viruses have a unique mechanism of replication. Their genome is a single strand RNA that has to be transcribed as soon as the virus enters the host in order to carry out viral replication. Viruses are intracellular parasites comprised of limited genetic information. As such, viruses must rely on either existing or modified cellular machineries for many of the steps involved in macromolecular synthesis. In this review of the currently published scientific literature, we detail the complexity and diversity of these hijacked cellular processes as it relates to viral transcription and/or replication in non-segmented negative strand RNA viruses of the order Mononegavirales. Identification of the interactions between virus proteins and host cellular proteins has important practical implications as they can provide new ways to combat viral infections via the control of host cellular components.

SeV reverse genetics originating from a pure academic need to settle long-held questions in the biology and pathogenicity of nonsegmented negative strand RNA viruses (Mononegavirales) is about to bear the impressive fruit of multipurpose cytoplasmic (non-integrating) RNA vectors. This book brings together in one source the SeV biology revealed by conventional approaches and reverse genetics, the methods to construct the first-generation SeV vector and to generate safer versions, and the applications in medical settings that have left or are about to leave the laboratory bench. The applications, which already are diverse and have high medical impact, include use as vaccine vectors against AIDS and respiratory virus infections, creation of BioKnife to resect malignant tumors, induction of “footprint (transgene) free” pluripotent stem cells, and gene therapy for peripheral arterial disease.
