This open access book is only an introduction to show that radiation and radioisotopes (RI) are premier tools to study living plant physiology which leads to new findings. Who had ever imagined that we could see water in a plant? Who had ever imagined that we could see ions moving toward roots in solution? Who had ever imagined that we could see invisible gas (CO2) fixation and movement in a plant? These studies demonstrated for the first time that water, ions and gas can be visualized in living plants, which could be hardly seen by anyone before. This publication summarizes the results obtained by Nakanishi’s lab in The Univ. of Tokyo, based on her original concept and her original tools or systems. It is useful for professional scientists, plant physiologist, and those studying plant imaging. The chapters demonstrates the innovative imaging work of the author, using radioactive tracers and neutron beam to follow the absorption and transport manner of water as well as major, minor, and trace elements in plants. Through these studies the author developed a real-time macroscopic and microscopic imaging system able to apply commercially available gamma- and beta-ray emitters. The real-time movement of the elements is now possible by using 14C, 18F, 22Na, 28Mg, 32P, 33P, 35S, 42K, 45Ca, 48V, 54Mn, 55Fe, 59Fe, 65Zn, 86Rb, 109Cd, and 137Cs. The imaging methods was applied to study the effect of 137Cs following 3/11 Fukushima Daiichi nuclear plant accident, which has revealed the movements of radiocesium in the contaminated sites.

The study of plant cell physiology is currently experiencing a profound transformation. Novel techniques allow dynamic in vivo imaging with subcellular resolution, covering a rapidly growing range of plant cell physiology. Several basic biological questions that have been inaccessible by the traditional combination of biochemical, physiological and cell biological approaches now see major progress. Instead of grinding up tissues, destroying their organisation, or describing cell- and tissue structure, without a measure for its function, novel imaging approaches can provide the critical link between localisation, function and dynamics. Thanks to a fast growing collection of available fluorescent protein variants and sensors, along with innovative new microscopy technologies and quantitative analysis tools, a wide range of plant biology can now be studied in vivo, including cell morphology & migration, protein localization, topology & movement, protein-protein interaction, organelle dynamics, as well as ion, ROS & redox dynamics. Within the cell, genetic targeting of fluorescent protein probes to different organelles and subcellular locations has started to reveal the stringently compartmentalized nature of cell physiology and its sophisticated spatiotemporal regulation in response to environmental stimuli. Most importantly, such cellular processes can be monitored in their natural 3D context, even in complex tissues and organs – a condition not easily met in studies on mammalian cells. Recent new insights into plant cell physiology by functional imaging have been largely driven by technological developments, such as the design of novel sensors, innovative microscopy & imaging techniques and the quantitative analysis of complex image data. Rapid further advances are expected which will require close interdisciplinary interaction of plant biologists with chemists, physicists, mathematicians and computer scientists. High-throughput approaches will become increasingly important, to fill genomic data with ‘life’ on the scale of cell physiology. If the vast body of information generated in the -omics era is to generate actual mechanistic understanding of how the live plant cell works, functional imaging has enormous potential to adopt the role of a versatile standard tool across plant biology and crop breeding. We welcome original research papers, methodological papers, reviews and mini reviews, with particular attention to contributions in which novel imaging techniques enhance our understanding of plant cell physiology and permits to answer questions that cannot be easily addressed with other techniques.

Plants and trees are among the most complex natural objects. Much work has been done attempting to model them, with varying degrees of success. In this book, we review the various approaches in computer graphics, which we categorize as rule-based, image-based, and sketch-based methods. We describe our approaches for modeling plants and trees using images. Image-based approaches have the distinct advantage that the resulting model inherits the realistic shape and complexity of a real plant or tree. We use different techniques for modeling plants (with relatively large leaves) and trees (with relatively small leaves).With plants, we model each leaf from images, while for trees, the leaves are only approximated due to their small size and large number. Both techniques start with the same initial step of structure from motion on multiple images of the plant or tree that is to be modeled. For our plant modeling system, because we need to model the individual leaves, these leaves need to be segmented out from the images. We designed our plant modeling system to be interactive, automating the process of shape recovery while relying on the user to provide simple hints on segmentation. Segmentation is performed in both image and 3D spaces, allowing the user to easily visualize its effect immediately. Using the segmented image and 3D data, the geometry of each leaf is then automatically recovered from the multiple views by fitting a deformable leaf model. Our system also allows the user to easily reconstruct branches in a similar manner. To model trees, because of the large leaf count, small image footprint, and widespread occlusions, we do not model the leaves exactly as we do for plants. Instead, we populate the tree with leaf replicas from segmented source images to reconstruct the overall tree shape. In addition, we use the shape patterns of visible branches to predict those of obscured branches. As a result, we are able to design our tree modeling system so as to minimize user intervention. We also handle the special case of modeling a tree from only a single image. Here, the user is required to draw strokes on the image to indicate the tree crown (so that the leaf region is approximately known) and to refine the recovery of branches. As before, we concatenate the shape patterns from a library to generate the 3D shape. To substantiate the effectiveness of our systems, we show realistic reconstructions of a variety of plants and trees from images. Finally, we offer our thoughts on improving our systems and on the remaining challenges associated with plant and tree modeling.
