The central theme of this book is that systems of cell-cell signalling via nerves, hormones, local mediators and growth factors are not distinct phenomena, but branches of one general mechanism. These topics therefore can and should be discussed in an integrated manner, and the division of cell signalling studies into separate pigeonholes such as neuroscience, endocrinology or cancer biology is unnecessary, if not counterproductive. I also believe it to be unfortunate that there is not a collective term to describe neurotransmitters, hormones, local mediators and growth factors, other than clumsy phrases such as "extracellular signal molecule". The lack of a short and distinctive word for these entities genuinely hampers people from thinking about them in an integrated way. Having decided that it was presumptuous to invent a new term, I have chosen in this book the term first messenger to cover all types of extracellular signal molecule, because of the widespread acceptance of the term second messenger to represent the intracellular signal molecules that are produced in response to many of them. I have given the book the title "biochemical messengers", which is a global term to cover both first and second messengers. The impetus for writing the book came, as must often be the case, when I had to put together a course on cell-cell signalling for biochemistry students at the University of Dundee.

This volume addresses a fundamental puzzle in biology and medicine, namely, how does tissue develop, repair and replace itself. The answer appears to lie in growth factors and their regulation. To thrive and survive we need growth factors and this book concentrates on two factors that are related to growth hormone. Growth hormone does not act directly on all tissues, but mediates many of its actions through the release of insulin-like growth factors from the liver. The growth factors were originally called somatomedins by McConaghey and Sledge (1), who discovered that they mediated growth-like effects of growth hormone. However, the factors were purified on the basis of their insulinomimetic actions on fat and muscle and it is their relationship to the insulin family of pep tides that now gives them their name (2,3) of insulin-like growth factors (IGFs). They mediate the actions of. growth hormone on the proteoglycan synthesis of cartilage and produce mitogenic effects in fibroblast cultures.

The biotechnological advances of recent years have put us on the brink of unprecedented gains in animal productivity. Manipulation of animal growth rate and composition of gain is now possible by a variety of techniques. Ex amples include ingestion of beta-adrenergic agonists, injection of somatotropin, castration, immunization, and gene insertion. Animal Growth Regulation ad dresses modem concepts of growth regulation with an emphasis on agricul turally important animals. This emphasis is not exclusive, as many situations exist in which the only information available was generated in other species, and this information has been included for the sake of clarity and completeness. However, because of the overall orientation of this volume, particular attention has been given to the regulation of skeletal muscle, adipose tissue, and bone growth. Certain hormones and growth factors have a profound influence on growth regulation and this basic physiological knowledge is being harnessed to maniplilate growth. Thus, considerable emphasis has been given to growth hor mone-somatomedinlinsulinlike growth factor regulation of cell and tissue growth. The involvement of peptides coded by protooncogenes and of negative growth regulators, such as transforming growth factor-l3, represents an emerging area of molecular biology wherein basic knowledge offers potential exploitation for growth manipulation.
