Em 1522, logo após finalizar a tradução e a edição do Novo Testamento em língua alemã, Lutero começou a traduzir o Antigo Testamento. Doze anos depois sua tradução completa da Bíblia seria publicada pela primeira vez. Os comentários de livros inteiros e passagens do Antigo Testamento traduzidos e editados com ineditismo para o português no presente volume pertencem justamente a esse período de intenso trabalho do professor, exegeta e tradutor bíblico. O envolvimento com o texto na função de professor perante seus alunos em sala de aula dava suporte ao labor na tradução e vice-versa, sendo nesta segunda tarefa assessorado por Filipe Melanchthon, Jorge Espalatino e Mateus Aurogalo, este professor de hebraico na universidade de Wittenberg. A atividade pública como docente, após o retorno do refúgio no Wartburgo, não se deu sem relativa tensão, pois do ponto de vista político era bem inapropriado permitir a um excomungado e banido ensinar em uma universidade sob jurisdição territorial. Isso fez, por exemplo, com que a preleção sobre Deuteronômio, que redundaria no comentário disponibilizado neste volume, fosse conduzida como um pequeno colóquio informal, ambientado no mosteiro agostiniano eremita, do qual Lutero era membro, e não na universidade. É possível observar como o andamento da preleção corresponde ao andamento da tradução, não apenas para o alemão, mas também para o latim, pois possibilidades de tradução para ambas as línguas são discutidas verso a verso com base em critérios filológicos. Suas fontes estavam mais referenciadas na exegese humanista do que na exegese medieval, principalmente pelo trabalho intenso com o texto original.
