oltage-gated calcium channels are essential mediators of a range of physiological functions, including the communication between nerve Vcells, the regulation of heart beat, muscle contraction, and secretion of hormones such as insulin. Consequently, these channels are critical phar macological targets in the treatment of a variety of disorders, such as epi lepsy, hypertension, and pain. Voltage-gated calcium channels have there fore been subject to intense study by numerous investigators over the past few decades, and an immense body of work has accumulated. In this book, we provide the first comprehensive overview of our current state of knowl edge concerning this exciting field of research. Leading off with a general review of calcium signaling and techniques to measure calcium channel ac tivity, the book delves into a provocative overview of the history of the cal cium channel field, contributed by one of the key pioneers in the field. Dr. Richard Tsien. This is followed by an in depth review of the biochemical and molecular biological characterization of calcium channel genes by Drs. Catterall and Snutch whose research has resulted in major advances in the calcium channel field. A number of chapters are dedicated towards various aspects of calcium channel structure and function, including channel gat ing, permeation, modulation and interactions with members of the exo- totic machinery—contributed by both established leaders and rising stars in the field.

This volume draws together studies on the diverse group of calcium-activated chloride channels (CaCCS) in one comprehensive format. The characteristics of CaCCs identified in a number of different systems are described by experts in the field. Channels discussed include those described in cardiac, neuronal, endothelial and epithelial systems, in addition to sections on the CaCC of Xenopus Oocytes and the newly identified family of CLCA proteins. Key Features * Includes description of CaCCs in excitable and non-excitable tissues * Reviews identification and cloning of the CLCA protein family * Contains section on CaCCs in Xenopus Oocytes

In the past few years, the scientific community has witnessed significant progress in the study of ion channels. Technological advancement in biophysics, molecular biology, and immunology has been greatly ac celerated, making it possible to conduct experiments which were deemed very difficult if not impossible in the past. For example, patch-clamp techniques can now be used to measure ionic currents generated by almost every type of cell, thereby allowing us to analyze whole-cell and single channel events. It is now possible to incorporate purified ion channel components into lipid bilayers to reconstitute an "excitable membrane." Gene cloning and monoclonal antibody techniques provide us with new approaches to the study of the molecular structure of ion channels. A variety of chemicals have now been found to interact with ion channels. One of the classical examples is represented by tetrodotoxin, a puffer fish poison, which was shown in the early 1960s to block the voltage-activated sodium channel in a highly specific and potent manner.
