This paper describes initial experiments to perform laser ablation mass spectrometry in real time on airborne microparticles. The microparticles are sampled directly from the air by a particle inlet system into the vacuum chamber of a mass spectrometer. An incoming particle is detected as it passes through two CW laser beams and a pulsed laser is triggered to intercept the particle for laser ablation/ionization in the mass spectrometer. The initial studies were made with an existing ion trap mass spectrometer with the particle sampling occurring at the center of the trap electrodes. Performance of the inlet system, particle detection, and preliminary results are described.

An important aspect of environmental atmospheric monitoring requires the characterization of airborne microparticles and aerosols. Unfortunately, traditional sample collection and handling techniques are prone to contamination and interference effects that can render an analysis invalid. These problems can be avoided by using real-time atmospheric sampling techniques followed by immediate mass spectrometric analysis. The former is achieved in these experiments via a two state differential pumping scheme that is attached directly to a commercially available quadruple ion trap mass spectrometer. Particles produced by an external particle generator enter the apparatus and immediately pass through two cw laser/fiberoptic based detectors positioned two centimeters apart. Timing electronics measure the time between detection events, estimate the particles arrival in the center of the ion trap and control the firing of a YAG laser. Ions produced when the UV laser light ablates the particle's surface are stored by the ion trap for mass analysis. Ion trap mass spectrometers have several advantages over conventional time-of-flight instruments. First, they are capable of MS/MS analysis by the collisional dissociation of a stored species, This permits complete chemical characterization of airborne samples. Second, ion traps are small and lend themselves to portable, field oriented applications.

It has been thirty years since one of the authors (EJD) began a collaboration with Professor Milton Kerker at Clarkson University in Potsdam, New York using light scattering methods to study aerosol processes. The development of a relatively short-lived commercial particle levitator based on a modification of the Millikan oil drop experiment attracted their attention and led the author to the study of single droplets and solid microparticles by levitation methods. The early work on measurements of droplet evaporation rates using light scattering techniques to determine the size slowly expanded and diversified as better instrumentation was developed, and faster computers made it possible to perform Mie theory light scattering calculations with ease. Several milestones can be identified in the progress of single microparticle studies. The first is the introduction of the electrodynamic balance, which provided more robust trapping of a particle. The electrodynamic levitator, which has played an important role in atomic and molecular ion spectroscopy, leading to the Nobel Prize in Physics in 1989 shared by Wolfgang Paul of Bonn University and Hans Dehmelt of the University of Washington, was easily adapted to trap microparticles. Simultaneously, improvements in detectors for acquiring and storing light scattering data and theoretical and experimental studies of the interesting optical properties of microspheres, especially the work on morphology dependent resonances by Arthur Ashkin at the Bell Laboratories, Richard Chang, from Yale University, and Tony Campillo from the Naval Research Laboratories in Washington D. C.

We are developing a method for the real-time analysis of airborne microparticles based on laser ablation mass spectroscopy. Airborne particles enter an ion trap mass spectrometer through a differentially-pumped inlet, are detected by light scattered from two CW laser beams, and sampled by a 10 ns excimer laser pulse at 308 nm as they pass through the center of the ion trap electrodes. After the laser pulse, the stored ions are separated by conventional ion trap methods. In this work thousands of positive and negative ion spectra were collected for eighteen different species: six bacteria, six pollen, and six particulate samples. The data were then averaged and analyzed using the Multivariate Patch Algorithm (MPA), a variant of traditional multivariate anal ysis. The MPA correctly identified all of the positive ion spectra and 17 of the 18 negative ion spectra. In addition, when the average positive and negative spectra were combined the MPA correctly identified all 18 species. Finally, the MPA is also able to identify the components of computer synthesized mixtures of the samples studied.

A new instrument, the NanoAPA, is being developed for these in-situ applications as an inexpensive, portable, and real-time instrument. The instrument is designed to perform ultrafine particle sizing and counting throught electronic control of a microfabricated device that charges sampled airborne particles with a corona ionizer and then incrementally size-separates, collects, and counts the number of particles in the aerosol.

Bioaerosol Detection Technologies written by Per Jonsson and has been published by Springer this book supported file pdf, txt, epub, kindle and other format this book has been release on 2014-07-31 with Science categories.

This book is intended to give technological background and practical examples, but also to give general insight into the on-going technology development in the area of biodetection. The content is therefore suitable for an array of stakeholders (decision makers, purchasing officers, etc.) and end-users of biodetection equipment within the areas of health, environment, safety and security, and military preparation. The book is divided into three sections. The first section discusses the fundamental physical and biological properties of bioaerosol's. The second section goes into more detail and discusses in-depth the most commonly used detection principles. The third section of the book is devoted to technologies that have been used in standoff applications. The last section of the book gives an overview of trends in bioaerosol detection. The reader of this book will gain knowledge about the different biodetection technologies and thus better judge their capabilities in relation to desired applications.
