This research monograph focuses on a biomolecular separation process that occurs within most cells. Two types of molecules, receptors and ligands, are separated and routed along different intracellular pathways; this is a critical step in the process of receptor-mediated endocytosis. The development of an understanding of the basic mechanisms of this separation process is presented, with an emphasis on discovering the fundamental and measurable parameters that influence the event. Mathematical models of sorting are evaluated to predict the range of possible outcomes. These are compared with a variety of experimental data on different receptor/ligand systems. In addition, the influence of the separation on overall receptor/ligand processing dynamics is discussed. The book is intended for both biomathematicians and biologists. It is not necessary to understand the details of the model equations and their solution in order to test the models experimentally. The analysis suggests experiments that might be done to further investigate the sorting process and the text details methods for analyzing these experimental results.

This volume focuses exclusively on those endocytic processes that sequester proteins by a selective, receptor-mediated mechanism. In such an endocytic process, cell surface receptors specifically bind protein ligands and localize them to specialized invaginations of the plasma membrane. These regions are coated pits, so named because they are lined on the cytoplasmic face with an ordered array of the protein, clathrin. It is this 'coat' which provides their characteristic electron microscopic image. Subsequently, these regions pinch off to form coated vesicles which rapidly lose their 'coat' and then fuse with other organelles or the plasma membrane. The hallmarks of ,this process are the specific receptors, coated pits, coated vesicles and an ordered sequence of transit events leading to delivery to selected locations. Receptor recognition, specific disposition of the endocytosed ligand and the existence of recep tor-ligand complexes at highest density in coated pits define the process as selective and concentrative. This topic has received ever increasing attention during the past few years. The evolving mechanisms are especially exciting because they come at a time when the conventional views based on thermodynamic arguments suggest that proteins should not be able to cross into the cell. Receptor-mediated endocytosis, however, reconciles the view that biological membranes should be impervious to macromolecules with the evidence that certain mac romolecules do gain entrance into the cell. During the last few years this field has been stimulated by studies on the uptake and processing of low density lipoproteins (LDL) by cells.

"The endocytic sorting of cell surface receptors is a crucial process for maintaining eukaryotic cell homeostasis and for organism's development and physiology. Several decades of research in this field have yielded the understanding of molecular processes governing endocytosis and postendocytic sorting of receptors. However the emergence of additional regulatory mechanisms for the individual receptors and discovery of new players of endocytic machinery provide new areas for investigation and research. This work provides understanding of the regulation of integrin [alpha]5[beta]1 and Tumor Necrosis Factor Receptor (TNFR) by endocytic sorting and ubiquitination, as well as the characterization of a novel endosomal sorting machinery.Our study of integrin [alpha]5[beta]1 trafficking has identified a dynamic mechanism for [alpha]5[beta]1 partitioning between the degradative and the recycling pathways, with a profound implication in cellular migration and invasion. The work presented here outlines the timecourse of integrin [alpha]5[beta]1 ubiquitination following ligand engagement, its deubiquitination by USP9x upon ligand dissociation in acidic endosomes, promoting the receptor resensitization and its cell surface return. The smaller fraction of integrins, that maintain their ubiquitination status, are targeted to the lysosomal degradation via the action of the ESCRT components (HD-PTP and UBAP1). Overall, this dynamic sorting of integrin receptors represents a novel mechanism of cell migration regulation by integrin recycling. Moreover, we have identified a key function for HD-PTP in the regulation of TNFR1 downstream signalling, affecting both NF-[kappa]B pathway and the induction of apoptosis following TNF[alpha] stimulation. TNFR1 endocytic sorting is poorly understood, and little is known about the role of ESCRT machinery in the sorting of TNFR1 or its downstream signalling. Our findings provide the first evidence for the negative regulation of TNFR1 signalling by ESCRT component HD-PTP, since its depletion leads to increased scaffolding of the TNFR1 signalling complex, accelerated NF-[kappa]B signalling and target gene expression. The impact of the endosomal sorting machinery of TNFR1 has potential implications in inflammation, apoptotic signalling, and cell survival. Finally, we identify and characterize novel binding partners of HD-PTP by mass spectrometry. Among these, we focus on Endofin, an endosomal protein capable of ubiquitin recognition, and essential for lysosomal sorting of polyubiquitinated cargo molecules. Endofin was proposed to act as an alternative ESCRT module, and we provide the evidence for its role in sorting of ubiquitinated receptors. Overall, these studies outline the key function of endocytic sorting mechanisms of cell surface receptors during cell migration and inflammation and propose a new ESCRT complex as a module involved in sorting of ubiquitinated molecules. Future studies are needed to examine the control of inflammatory response by ESCRT machinery and the function of ESCRTs in carcinogenesis. " --
