This Volume contains the papers presented by twenty-eight invited speakers at the symposium entitled, "Genetic Manipulation of Woody Plants," held at Michigan State University, East Lansing, Michigan, from June 21-25, 1987. Also included are abstracts of contributed poster papers presented during the meeting. That the molecular biology of woody plants is a rapidly expanding field is attested to by the large attendance and high level of enthusiasm generated at the conference. Leading scientists from throughout the world discussed challenging problems and presented new insights into the devel opment of in vitro culture systems, techniques for DNA analysis and manipulation, gene vector systems, and experimental systems that will lead to a clearer understanding of gene expression and regulation for woody plant species. Soil acidity is a major limitation to crop production in many parts of the world. Plant growth inhibition results from a combination of factors, including aluminum, manganese, and hydrogen ion toxicities and deficiencies of essential elements, particularly calcium, magnesium, phosphorus, and molybdenum. Agricultural management practices and acid precipitation have increased acid inputs into the ecosystem and heightened concern about soil acidity problems. While application of lime has proved to be effective in ameliorating surface soil acidity in many areas, significant soil acidity problems still exist. Scientists from Alberta, Canada, recognized the need to provide a forum for researchers from different disciplines to exchange information and ideas on solving problems of plant growth in acid soils. As a result of their efforts, the First International Symposium on Plant-Soil Interactions at Low pH was held at Grande Prairie, Alberta, Canada, in July 1987. In many acid soil areas, liming materials are not readily available, the cost may be prohibitive, or subsoil acidity cannot be corrected by surface application of lime. New management approaches involving both the plant and the soil are needed in these situations. Progress has been made in the selection and breeding of acid-tolerant plants. However, continued progress will be limited by our lack of understanding of the physiological and biochemical basis of differential acidity tolerance among plants.

This publication, a compilation of 28 technical articles on various aspects of forest nursery management in western North America, consists of two sections. The first 10 papers comprise the Target Seedling Symposium, and discuss the latest methods of describing and measuring the ideal seedling for reforestation purposes. Considerable attention is paid to physiological tests such as root growth potential, hydraulic conductivity, cold-hardiness, mineral nutrition, seedling moisture status, and mitotic index. Chlorophyll fluorescence (Fvar), stress-induced volatile emissions (SIVE), and electrolyte conductivity (EC) are also discussed. In the last decade, enormous progress has been made on the physiology of plant roots, including on a wide range of molecular aspects. Much of that progress has been captured in the chapters of this book. Breakthroughs have been made possible through integration of molecular and whole-plant aspects. The classical boundaries between physiology, biochemistry and molecular biology have vanished. There has been a strong focus on a limited number of model species, including Arabidopsis thaliana. That focus has allowed greater insight into the significance of specific genes for plant development and functioning. However, many species are very different from A. thaliana, in that they are mycorrhizal, develop a symbiosis with N2-fixing microsymbionts, or have other specialised root structures. Also, some have a much greater capacity to resist extreme environments, such as soil acidity, salinity, flooding or heavy-metal toxicities, due to specific adaptations. Research on species other than A. thaliana is therefore pivotal, to develop new knowledge in plant sciences in a comprehensive manner. This fundamental new knowledge can be the basis for important applications in, e.g., agriculture and plant conservation. Although significant progress has been made, much remains to be learnt. It is envisaged that discoveries made in the recent past will likely lead to major breakthroughs in the next decade.
