Over the past 50 years many in vitro and in vivo drug response assay systems have been developed to determine the potential - tivity of chemotherapy agents. The idea was to eliminate ineffective agents and unnecessary toxic treatment while selecting drugs active in vitro or in the mouse model that might increase the probability of response in the patient. None of these test models, however, achieved routine clinical application in the past. This might be at least in part - lated to large discrepancies that were described between the s- cess rate of the assay systems and the clinical benefit in cancer - tients. The heterogeneity of chemosensitivity that exists between different tumors as well as between individual tumor lesions may be one explanation for these findings. Furthermore, different assay end points such as proliferation, metabolism, and vitality were - veloped to evaluate the effects of cytostatic drugs on tumor cells, and these might be related to the differing results. However, knowledge about procedures for assay-assisted treatment selection has increased rapidly within the past few years, and several studies suggest that test-directed chemotherapy selection now may - prove response rates and survival in various types of tumors. The International Society for Chemosensitivity Testing in - cology (ISCO) was founded to promote, coordinate, and improve clinical and laboratory research in the field of predictive drug te- ing in human tumor cells.

This text describes a comprehensive adjoint sensitivity analysis methodology (C-ASAM), developed by the author, enabling the efficient and exact computation of arbitrarily high-order functional derivatives of model responses to model parameters in large-scale systems. The model’s responses can be either scalar-valued functionals of the model’s parameters and state variables (as customarily encountered, e.g., in optimization problems) or general function-valued responses, which are often of interest but are currently not amenable to efficient sensitivity analysis. The C-ASAM framework is set in linearly increasing Hilbert spaces, each of state-function-dimensionality, as opposed to exponentially increasing parameter-dimensional spaces, thereby breaking the so-called “curse of dimensionality” in sensitivity and uncertainty analysis. The C-ASAM applies to any model; the larger the number of model parameters, the more efficient the C-ASAM becomes for computing arbitrarily high-order response sensitivities. The text includes illustrative paradigm problems which are fully worked-out to enable the thorough understanding of the C-ASAM’s principles and their practical application. The book will be helpful to those working in the fields of sensitivity analysis, uncertainty quantification, model validation, optimization, data assimilation, model calibration, sensor fusion, reduced-order modelling, inverse problems and predictive modelling. It serves as a textbook or as supplementary reading for graduate course on these topics, in academic departments in the natural, biological, and physical sciences and engineering. This Volume Three, the third of three, covers systems that are nonlinear in the state variables, model parameters and associated responses. The selected illustrative paradigm problems share these general characteristics. A separate Volume One covers systems that are linear in the state variables.

The U.S. Environmental Protection Agency has recommended that states develop Pesticide Management Plans for four agricultural chemicals - alachlor, atrazine, metolachlor, and simazine - used in Utah as herbicides in the production of corn and sorghum, and to control weeds and undesired vegetation (such as along right-of-ways or utility substations). This report and accompanying maps are intended to be used as part of these Pesticide Management Plans to provide local, state, and federal government agencies and agricultural pesticide users with a base of information concerning sensitivity and vulnerability of ground water in the basin-fill aquifer (bedrock is not evaluated) to agricultural pesticides in the central Virgin River basin, Washington and Iron Counties, Utah. We used existing data to produce pesticide sensitivity and vulnerability maps by applying an attribute ranking system specifically tailored to the western United States using Geographic Information System analysis methods. 24 pages + 2 plates
