A number of analytical and preparative problems of complex biological mixtures, requiring speed and mildness of handling, large capacity, and high resolution, can be advantageously attacked by the stable-flow free boundary (STAFLO) method. Experimental and theoretical aspects of STAFLO migration, fractionation, and concentration are analyzed, with particular reference to cell mixtures. Combination gradient profiles of composition, conductivity, pH, et cetera, can be established almost arbitrarily in a steady-state manner, without the usual gradient mixing devices. The solution to the basic problem of stabilizing flows, without the use of an anticonvective supporting medium, is described in detail. Migration principles, especially for sedimentation and electrophoresis are classified as rate, or equilibrium, and the known and unknown elements of several examined. A simplified Stokes-type treatment is sufficient for semiquantitative description of individual cell sedimentation under 1 g, including calculation of resolution and separation time. A kind of experimental cell behavior called bombing'' is discussed in terms of weak intercellular interaction. Analysis of STAFLO-electrophoresis and STAFLO-sedimentation-electrophoresis in terms of flow, heat production, and other parameters, indicates the basis for preparative cell fractionation, with steady-state times generally of from 2 to 60 minutes. An immediate possibility from such studies is the characterization of cells in mixtures according to sedimentation and electrophoretic mobilities; these latter must eventually be more unequivocally related to cell density'', size'', surface charge and other physical properties. Such relations require more detailed knowledge of cell and solution theory and perhaps better definitions of these properties when applied to cells. As an analytical example of instantaneous determination of a time-dependent transport property, an electrophoretic mobility of a dye is estimated from the slope of a steadystate STAFLO-electrophoresis, migration pattern. 'lbere is much discussion today concerning "Bioengineering" (or "Biomedical Engineering"). It is not exactly clear what these names signify, particularly in chemical engineering. Some have suggested retreading the old war horse "Biochemical Engineering" (or was it "Biomedical Chemical Engineering). In an effort to demonstrate the on-going activities of chemical engineers in the life science area, we accepted the invitation of the Industrial and Engineering Division of the American Chemical Society to organize the 33rd Annual Chemical Engineering Symposium. We decided to call the symposium, Chemical Engineering in Medicine and Biology, and hence avoided the problem of having to decide which "bio" prefix to use. Many chemical engineers in the academic and industrial world were contacted. From these contacts and a good deal of publicity arose the Symposium. The two-day meeting was held at the University of Cincinnati in the Losantiville Room of the Student Union Building on October 20-21, 1966. Twenty-one papers were presented on topics relating chemical engineering to medicine and biology. Tile papers were representation al of the scope of the activities across the country with presenters coming from Washington, California, Massachusetts, New York, South Carolina, Wisconsin, Iowa, Pennsylvania, Michigan, Indiana and Texas. TOpics ranged over blood flow properties, diffusion in blood phenomena, ix INTRODUCTION X mass transfer in the eye, artificial kidney analysis, separation of bacteria by ion exchange, mathematical modeling of drug distribution, carbon dioxide respiration, photosynthetic kinetics, water in frozen tissues, electrophoretic separation of proteins, and outerspace re search on life support systems.

Presently, the need for methods involving separation, identification, and characterization of different kinds of cells is amply realized among immu nologists, hematologists, cell biologists, clinical pathologists, and cancer researchers. Unless cells exhibiting different functions and stages of differ entiation are separated from one another, it will be exceedingly difficult to study some of the molecular mechanisms involved in cell recognition, specialization, interactions, cytotoxicity, and transformation. Clinical diag nosis of diseased states and use of isolated cells for therapeutic (e. g. , immunotherapy) or survival (e. g. , transfusion) purposes are some of the pressing areas where immediate practical benefits can be obtained by applying cell separation techniques. However, the development of such useful methods is still in its infancy. A number of good techniques exist based either on the physical or biological properties of the cells, and these have produced some valuable results. Still others are to be discovered. Therefore, the purpose of this open-end treatise is to acquaint the reader with some of the basic principles, instrumentation, and procedures pres ently in practice at various laboratories around the world and to present some typical applications of each technique to particular biological prob lems. To this end, I was fortunate to obtain the contribution of certain leading scientists in the field of cell separation, people who in their pioneer ing work have struggled with the particular problems involved in separating living cells and in some way have won.
