Stevioside is one of the naturally occurring sweeteners, belonging to the diterpene glycoside family, which can be widely applied in food, drinks, medicine and consumer chemicals. Stevia Rebaudiana is a perennial herbaceous plant, part of the Asteraceae family and native to Paraguay. It has been cultivated in China and Southeast Asia for several decades and more recently in France, in the Occitanie region. More commonly known as “Stevia”, it is a new source of natural sweetener since it contains steviol glycosides with a sweet taste. These molecules have a sweetening power 100-300 times greater than sucrose and they are calorie-free. The major steviol glycosides in the plant are Stevioside (Stv) and Rebaudioside A (Reb A). The objective of this thesis is the development of unit operations with controlled properties of crystallization and drying in order to obtain purified powders, from juices extracted from organic stevia leaves. This project was carried out in collaboration with the start-up Oviatis, the Occitanie region and École des Mines d'Albi.

Stevioside can be obtained in Stevia rebaudiana leaves , which originated from Paraguay. It has been planted across Asia, Europe and Canada. It is the source of the highly sweet ent-kaurenoid diterpene glycosides, rebaudiana A, stevioside and other several sweet analogs. Stevioside is 300 times sweeter than sucrose and has lately gained importance as a natural non-caloric sweetener. The commercial exploration of S. rebaudiana has become stronger since 70's due to the Japanese researchers developed extraction and purification process of sweetener. In S. rebaudiana, the ratio of stevioside to rebaudiaoside-A is 2:1. For its dominance, stevioside imparts a bitter after-taste characteristic to the crude extracts. The main purpose of this to study the effect of solvent concentration, extraction time and the ratio leaves to volume of solvent on the stevioside extraction. The soxhlet extractor was used in this study. Analysis of stevioside was carried out by anthrone reaction. From the results, extraction using 70% ethanol resulted in the highest stevioside yield at 12 mg/mL. Meanwhile, the highest stevioside at 16.5 mg/mL yield can be obtained during 24 hours extraction time. For the mass leaves to volume of solvent, 0.09g/mL of the mass of the leaves gave the highest stevioside yield at 32.5 mg/mL.

Stevia rebaudiana is a sweet, green leaves that contain high amount of sweetener but has no calories. Inside stevia rebaudiana leaves, there are many sweet components includes stevioside, rebaudioside A, B, D, E and dulcoside. Among these sweet components, stevioside is the most abundant component. Stevioside is a natural sweetener and 300 times sweeter than sucrose. It is an alternative to sugar, that suitable for people who suffer from diabetes and hypertension because it has a negligible effect on blood glucose. In this present study, stevioside was extracted using an ultrasonic extraction method. They were 3 parameters that have been studied. The screening of the best extraction solvents (distilled water, ethanol 95% and 70% ethanol aqueous) were firstly investigated before designing the experiment by using one-factor-at-one time method. The 3 parameters involved included the ratio of leaves/solvent, extraction time and the ultrasonic frequency. Based on the result obtained, distilled water is the best extraction solvent with the ratio of leaves to solvent at 4:30 (w/v), extraction time of 20 minutes and the sonicator frequency at 20 kHz. It can be concluded that by using suitable types of solvent ratio, extraction time and sonicator frequency, the highest yield of stevioside have been obtained.
