The first concern of scientists who are interested in synthetic polymers has always been, and still is: How are they synthesized? But right after this comes the question: What have I made, and for what is it good? This leads to the important topic of the structure-property relations to which this book is devoted. Polymers are very large and very complicated systems; their character ization has to begin with the chemical composition, configuration, and con formation of the individual molecule. The first chapter is devoted to this broad objective. Phase transition of polymer molecules in solution, one of the most important topics in polymer and biomolecular physics, is dominated by the net interactions between polymer and solvents. By controlling the thermodynamic and kinetic processes of polymer solution phase separation with temperature-shifting or solvent-shifting, different morphologies of the separated polymer phase from solution can be obtained, with applications including polymer nanoparticle fabrication, polymer crystallization and protein folding. Polymer nanoparticles of high purity, controlled size and narrow size distribution are desired for biomedical applications. One of the promising methods to produce nanoparticles is the solvent-shifting process, in which a non-solvent is added to a pre-synthesized polymer in solution to precipitate polymer nanoparticles. The mechanism of the formation of these polymer nanoparticles is not well understood, particularly the initial nucleation process in which the polymer chain undergoes comprehensive molecular conformation changes. In this project, we study the coil-to-globule transition induced by adding water (non-solvent) into Poly(methyl methacrylate) (PMMA) solution in Tetrahy-drofuran (good solvent). Formation of polymer multi-chain clusters and the coil-to-globule transition of these clusters are observed. An intermediate state of multi-chain clusters is observed during the transition with lifetimes of days to months, depending on the concentration of non-solvent (water). The collapsed multi-chain clusters are stable in the solution after the transition with no further growth in time. This aggregation and collapse transition of polymer chains is compared to the traditional polymer single chain two-step coil-to-globule transition in dilute solution induced by temperature-shifting, during which a sausage-shaped intermediate state has been observed with lifetimes of minutes to hours. The formation of stable multi-chain clusters in solution is discussed and compared to a well-known surfactant free microemulsion (SFME) system in small molecule ternary systems. The formation of small molecular nm-size droplets internary systems is explained using two competing mechanisms. Entropy change favors smaller size clusters and, as the clusters get smaller, while the average distance between particles shrink and leading to an opposing hydration force. In the formation of multi-chain cluster, no such hydration force existence with the distance of hundreds of nanometers between clusters. One possible mechanism is that the competition between interfacial energy and entropic term gives a specific PMMA globule radius. In order to understand the real interfacial energy at the interface between PMMA phase and the co-solvent phase, detailed structure at the interface is investigated using Sum Frequency Generation technique. A condensation of ordered THF molecules at the interface was observed. Further measurement of the interfacial energy and investigation of real structure at the interface are necessary to explain the formation of stable multi-chain clusters with specific size. An understanding of the nanoparticle formation process leads to the design of a continuous membrane-based process to produce pure PMMA nanoparticles with controlled size and narrow size distribution. The size of these nanoparticles varies with the concentration of the PMMA feed and the amount of water diffusing across the membrane. The size distribution of PMMA particles produced in acontinuous flow membrane cell is much narrower than those produced by drop-wisewater addition or batch dialysis precipitation of PMMA particles. A continuous production of polymer nanoparticles of high purity and narrow dize distribution is important for biomedical applications such as delivering therapeutics.
