Knowledge of the extracellular matrix (ECM) is essential to understand cellular differentiation, tissue development, and tissue remodeling. This volume of the series “Biology of Extracellular Matrix” provides a timely overview of the structure, regulation, and function of the major macromolecules that make up the extracellular matrix. It covers topics such as collagen types and assembly of collagen-containing suprastructures, basement membrane, fibronectin and other cell-adhesive glycoproteins, proteoglycans, microfibrils, elastin, fibulins and matricellular proteins, such as thrombospondin. It also explores the concept that ECM components together with their cell surface receptors can be viewed as intricate nano-devices that allow cells to physically organize their 3-D-environment. Further, the role of the ECM in human disease and pathogenesis is discussed as well as the use of model organisms in elucidating ECM function.

In the ten-year interval since the first edition of this volume went to press, our knowledge of extracellular matrix (ECM) function and structure has enor mously increased. Extracellular matrix and cell-matrix interaction are now routine topics in the meetings and annual reviews sponsored by cell biology societies. Research in molecular biology has so advanced the number of known matrix molecules and the topic of gene structure and regulation that we won dered how best to incorporate the new material. For example, we deliberated over the inclusion of chapters on molecular genetics. We decided that with judicious editing we could present the recent findings in molecular biology within the same cell biology framework that was used for the first edition, using three broad headings: what is extracellular matrix, how is it made, and what does it do for cells? Maintaining control over the review of literature on the subject of ECM was not always an easy task, but we felt it was essential to production of a highly readable volume, one compact enough to serve the the student as an introduction and the investigator as a quick update on graduate the important recent discoveries. The workings of the suitable environment for cells—called the extracellular matrix (ECM) and ground regulation—has occupied the European medical tradition since the early part of the 20th century. As it has become more clear that the origin of disease and its first signals register in the connective tissue, or myofascia, cellular pathologists and biochemists have sought to circumscribe networks of cell communication and microcirculation in the ECM. Alfred Pischinger (1899-1982) continued this line of work by further studying, in work published from 1926 through the late seventies, the connections of the ECM to the hormonal and autonomic systems. In the last twenty years Professor and Doctor of Natural Sciences Hartmut Heine and his colleagues have carried on Pischinger’s work, here summarized in one volume. Part One encompasses theoretical underpinnings; Parts Two and Three address applications and directions for further research. This updated English-language translation not only is an account of the work of Pischinger’s successors—Heine, Otto Bergsmann, and Felix Perger, (the three editors of this volume) and their many colleagues—but notes the positive development of complementary therapies based on this understanding of histology. Acupuncture is referenced directly. Both in Europe and the States the work of manual therapists, including Rolfers, cranio- sacral therapists, and other somatic disciplines have been informed for many years by Pischinger’s outsider model of how changes in the EMC register in the central nervous system and the brain, and are conveyed back to the periphery and connected organs. Heine’s exciting recent work shows that the regulation and construction of the ECM have relationships to cybernetic non-linear systems and phase transitions.

The extracellular matrix (ECM) is the focus of much interest in biology and bioengineering. Increasing understanding of the influence of the ECM on cell behaviour has led to the exciting possibilities of tissue engineering. Aside from new therapeutic tools, understanding the ECM is of course fundamental to basic cell biology research. Mimicking the Extracellular Matrix approaches this topic from both basic science and practical engineering perspectives. Seven topics are approached each in a pair of chapters, one with a biological approach and its partner with a bioengineering approach. Topics include the mechanical properties of the ECM, which outlines current knowledge of the ECM physical structure and reviewing state-of-the-art strategies to mimic its native microenvironments. The organisational characteristics of the ECM form the focus of another pair of chapters, where the collagen triple helix is discussed, followed by a review of advances in artificial reproduction of well-ordered systems using self-assembling peptides, or peptide amphiphiles. The balanced approach of this text gives it a broad appeal to those interested in the ECM from a range of backgrounds and disciplines. Suitable for undergraduates, postgraduates, and academics, this text aims to unify the current knowledge of ECM biology and matrix-mimicking biomaterials.

Studies of the extracellular matrix (the material that surrounds cells and tissues) span much of modern biological research. In addition to providing structure to organisms the extracellular matrix is crucial in determining cell growth, how the cell responds during injury and development and what the cell makes. Changes in the extracellular matrix are important components in both inherited and acquired diseases such as diabetes and cancer. This important volume brings together undera single cover the majority of techniques currently employed in extracellular matrix research and will serve as an indispensable resource for researchers and clinicians in this field.

Structure and Function of the Extracellular Matrix: A Multiscale Quantitative Approach introduces biomechanics and biophysics with applications to understand the biological function of the extracellular matrix in health and disease. A general multiscale approach is followed by investigating behavior from the scale of single molecules, through fibrils and fibers, to tissues of various organ systems. Through mathematical models and structural information, quantitative description of the extracellular matrix function is derived with tissue specific details. The book introduces the properties and organization of extracellular matrix components and quantitative models of the matrix, and guides the reader through predicting functional properties. This book integrates evolutionary biology with multiscale structure to quantitatively understand the function of the extracellular matrix. This approach allows a fresh look into normal functioning as well as the pathological alterations of the extracellular matrix. Professor Suki’s book is written to be useful to undergraduates, graduate students, and researchers interested in the quantitative aspects of the extracellular matrix. Researchers working in mechanotransduction, respiratory and cardiovascular mechanics, and multiscale biomechanics of tendon, cartilage, skin, and bone may also be interested in this book. Examines the evolutionary origins and consequences of the extracellular matrix Delivers the first book to quantitatively treat the extracellular matrix as a multiscale system Presents problems and a set of computational laboratory projects in various chapters to aid teaching and learning Provides an introduction to the properties and organization of the extracellular matrix components

It is now widely accepted that much of the dynamic function of cells and tissues is regulated from outside the cell by the extracellular matrix. In ad- tion to its conventional role in providing a scaffold for building tissues, the extracellular matrix acts as a directional highway for cellular movement and provides instructional information for promoting survival, proliferation, and differentiation. Indeed, the extracellular matrix is beginning to take a starring role in the choreography of cell and tissue function. The diverse roles of the extracellular matrix are reflected in its highly complicated structure, consisting of an ever increasing number of components. Yet the mechanisms of extracellular matrix assembly and how they influences cell behavior are only just beginning to be understood. In order to solve these problems new methodologies are, of necessity, being developed. Many of these technologies are highly sophisticated and are currently available only in a ha- ful of laboratories. However, we believe that they can readily be transported and established by other researchers. Thus, the purpose of Extracellular Matrix Protocols is to present some of these complicated techniques in a style that is relatively easy to reproduce.

Extracellular Matrix contains the proceedings of the symposium ""The Extracellular Matrix,"" sponsored by the Michigan Molecular Institute and held in Midland, Michigan, on June 28-July 2, 1982. The papers explore the role played by the extracellular matrix (ECM) in the physiology of a cell, particularly in the regulation of cellular phenotypes, differentiation, and proliferation. The progress made in isolating and defining the chemistry and functional interactions of the ECM components is discussed, along with the biology of the ECM. This book is comprised of 52 chapters and begins with an introduction to the ECM, with emphasis on the question of whether the malignant process can be defined in a cell culture model, and in particular, whether the pericellular matrix is characteristically altered in cancer. The discussion then turns to the structure of the heparan sulfate proteoglycans and the molecular mechanisms responsible for the association of these molecules with the surfaces of cultured cells. Subsequent chapters focus on the chemistry of ECM components such as collagen, glycosaminoglycans, and adhesive glycoproteins, along with their functional interactions, biosynthesis, turnover, and degradation. The final section is devoted to the diseased states of ECM. This monograph should serve as a valuable reference for biochemists as well as undergraduate and graduate students of biochemistry.
