The Nucleus Accumbens (NAcb), part of the ventral striatum in the basal ganglia, processes afferent input from many of the cognitive and limbic areas of the brain, including the prefrontal cortex (PFC), hippocampus (HC), amygdala, and thalamus. Altered processing in this region of the brain has been hypothesized to underlie disorders such as schizophrenia and addiction. The central question of this thesis is how the inputs to the NAcb are processed by the major cell in the NAcb, the medium spiny projection neurons (MSPs). In order to investigate this problem, we developed a computational model of the cell and examined its response to afferent inputs, as well as recording responses of these cells to stimulation of afferent inputs in the awake rat. The model neuron, constructed in NEURON, was tuned by adjusting maximal current conductances in each compartment until the model cell closely matched whole cell recordings from an adult rat NAcb slice preparation. Synaptic inputs in the range of 1000-1300 Hz are required to maintain an "up" state in the model. As NMDA:AMPA current ratios were increased, the membrane showed an increase in hysteresis of "up" and "down" state dwell times, however intrinsic bistability was not observed. Altering the NMDA:AMPA ratio had a profound effect on processing of afferent input, including the ability to entrain to oscillations in afferent input in the theta range (4-12 Hz). To investigate the hypothesis that PFC inputs to the NAcb are gated by HC inputs, we extended previous studies in anesthetized rats by comparing the response to various combinations of PFC and HC stimulation in the awake and the isoflurane anesthetized, chronically implanted rats. Contrary to previous findings, we found that summation of afferent input in the NAcb is sublinear, with only a minority of neurons demonstrating gating of the PFC inputs via the HC in the awake or the anesthetized animal. These results suggest that sublinear integration of afferent inputs is the dominant profile in the response of NAcb cells in the awake animal.
