Toxicity Assessment Alternatives: Methods, Issues, Opportunities contains a broad array of critical surveys, contributed by active and respected investigators, describing their research and offering updates on toxicity assessment alternatives, directions determined by current and future grant programs, opportunities for mechanistically based test methods to detect endocrine disruptor activity, the use of alternatives in the Department of Defense hazard assessment initiatives, and the issues and opportunities for validation and regulatory acceptance. Several of these advances make use oftransgenic models that reduce the time and cost of carcinogenicity testing. Others use tissue cultures for the assessment of endocrine disrupting chemicals. Cultures of human epidermal keratinocytes are applicable as models for sulfur mustard lesions, and in vitro protein denaturation is used as a chemical test for assessing the ocular and dermal irritation potential of cosmetic prod ucts. Molecular modeling is applied to explaining chemical toxicity. Commercially developed assay systems have undergone extensive evaluation by their manufacturers. Some of these await external valida tion, and others await acceptance by North American and European regulatory agencies. Toxicity Assessment Alternatives: Methods, Issues, Opportunities provides information from members of the scientific and regulatory communities on what has been achieved and what has been accepted in alternatives to animal testing.

Toxicity testing in laboratory animals provides much of the information used by the Environmental Protection Agency (EPA) to assess the hazards and risks associated with exposure to environmental agents that might harm public health or the environment. The data are used to establish maximum acceptable concentrations of environmental agents in drinking water, set permissible limits of exposure of workers, define labeling requirements, establish tolerances for pesticides residues on food, and set other kinds of limits on the basis of risk assessment. Because the number of regulations that require toxicity testing is growing, EPA called for a comprehensive review of established and emerging toxicity-testing methods and strategies. This interim report reviews current toxicity-testing methods and strategies and near-term improvements in toxicity-testing approaches proposed by EPA and others. It identifies several recurring themes and questions in the various reports reviewed. The final report will present a long-range vision and strategic plan to advance the practices of toxicity testing and human health assessment of environmental contaminants.

Wildlife Toxicity Assessments for Chemicals of Military Concern is a compendium of chemical-specific toxicity information with discussions on the rationale and development of Wildlife Toxicity Reference Values (TRVs) intended for use on terrestrial wildlife for risk assessment applications. Substances covered include military-related chemicals including explosives, propellants, pesticides and metals. Wildlife Toxicity Assessments for Chemicals of Military Concern is a much-needed resource designed to meet the needs of those seeking toxicological information for ecological risk assessment purposes. Each chapter targets a specific chemical and considers the current knowledge of the toxicological impacts of chemicals to terrestrial wildlife including mammalian, avian, amphibian and reptilian species. Provides detailed information on how Wildlife Toxicity Values (TRVs) for military chemicals of concern are derived and evaluated. Covers wildlife toxicity assessments of explosives, metals and environmental chemicals. Compiles relevant information on the environmental effects of chemicals on wildlife in relation to public and environmental health.

This book collects protocols from different areas of knowledge to assist in the identification of toxic effects exerted by different xenobiotics. At the same time as classical techniques are presented, modern techniques with alternative models to the use of animals are also presented. Given the ever-increasing exposure to different compounds and their effects on population health, the assessment of multiple endpoints is of utmost importance for better risk assessment, and this collection addresses that need. Written for the highly successful Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Toxicity Assessment: Methods and Protocols aims to serve researchers in this vast field of science as they seek to better understand the mechanisms of action of different xenobiotics.

Toxicogenomics is the integration of genomics to toxicology. This technology is a powerful tool for collecting information from a large number of biological samples simultaneously and thus it is very useful for large-scale screening of potential toxicants. Toxicogenomics: A Powerful Tool For Toxicity Assessment provides up-to-date state-of-the-art information presented by the recognized experts, and is therefore an authoritative source of current knowledge in this field of research. The potential link between toxicology, genetics and human diseases makes this book very useful to investigators in many and varied disciplines of science and toxicology. Topics covered include: mechanistic toxicogenomics analysis and interpretation of toxicogenomic data principles of data mining in toxicogenomics design issues in toxicogenomics studies sources of variability in toxicogenomic assays Escherichia coli stress response as a tool for detection of toxicity toxicogenomics as a tool to assess immunotoxicity toxicogenomics and ecogenomics for studying endocrine disruption and basic biology use of toxicogenomics as an early predictive tool for hepatotoxicity nutrigenomics: the application of genomic signatures in nutrition-related research application of toxicogenomics in drug discovery potential uses of toxicogenomic biomarkers in occupational health and risk assessment usefulness of toxicogenomics in the regulatory environment perspectives on toxicogenomics at the US Environmental Protection Agency Toxicogenomics: A Powerful Tool For Toxicity Assessment is an essential resource for research scientists currently engaged in toxicogenomics, and will also be of interest to researchers working in toxicology, genetics, medicine, pharmacology, and food sciences, and to regulators and risk assessors of drug, food, environmental and agricultural products.

Sediment Toxicity Assessment provides the latest information regarding how to evaluate sediment contamination and its effects on aquatic ecosystems. It presents an integrated ecosystem approach by detailing effective assessment methods, considerations, and effects to each major component of marine and freshwater systems, including the benthos, plankton, and fish communities. The approaches emphasize defining habitat conditions (physical and chemical), toxicant bioavailability, factors influencing toxicity (lab and field), biomarkers, acute and chronic toxicity, study design, collection methods, and EPA management strategies. The book also explains how to integrate the assessments. Sediment Toxicity Assessment will be useful to to all environmental managers, environmental scientists, ecotoxicologists, environmental regulators, aquatic ecologists, environmental contractors and consultants, instructors, students, conservation commissions, and environmental activist organizations.
