Alcoholism is a uniquely human condition. Although some forms of alcohol dependence can be induced experimentally in a variety of laboratory animals, the complete spectrum of alcoholism with all of its physical, psychological, and social implications occurs only in man. The special quality of this relationship becomes more significant when one considers that the manifestations of most physical disease syndromes in animals and man are more similar than they are different. The uniqueness of alcoholism lies in the fact that it is one of the few physical diseases which reflects at all levels the problems of individuals coping with the complexities of human society. In order to present a more coherent picture of these complex relationships, we have attempted to impose a logical sequence upon the material. This sequence lies along a dual parameter-from the physical to the social and from the theor etical to the empirical. Consequently, it was natural for the first volume in this series to deal with biochemistry, the most basic and physical aspect of the inter action of alcohol and man. It is equally natural for this, the second volume, to deal with physiology and behavior, for these levels of phenomenology-partic ularly the latter-are already more empirical and psychological in their mani festations. Finally, the third volume, clinical pathology, describes the disease itself, with all of the medical and social implications carried in the word "alcoholism.

Alcohol abuse throughout the world is associated with serious social and medical implications. Problems such as intoxication, tol erance, and development of physical dependence have been well recognized. The central nervous system and the liver are especially affected. There is little doubt that alcohol abuse can result in organ damage, which in turn leads to deleterious health consequences to the individual. Understanding ethanol action presents a special and functional diver challenge because of its molecular simplicity sity. In fact, the ability for alcohol to disrupt cellular function is at tributed to its cellular injury without regard to an apparent specific mechanism of action. Nevertheless, the key to an effective treatment to this problem is through research into understanding the mechan isms underlying how ethanol interacts with cells and membranes. This book is the result of a cooperative effort among scientists from many nations who met in a symposium in Taipei, Taiwan, ROC, July 1988. The focus of this book is on experimental ap proaches to better understand the molecular mechanisms of ethanol on the biological system. These recent advances in the examination of the alcohol effects on cellular function are divided into four sec tions. The first section addresses specific actions of ethanol on the central nervous system. The second section is directed to the use of cell cuI tures in ethanol research and the usefulness of cell cultures in examination of the effects of ethanol in vitro.

This is the first book about both normal development of the nervous system and how early exposure to alcohol and nicotine interferes with this development. The developing nervous system is highly dynamic and vulnerable to genetic and epigenetic factors that can be additive or synergistic. Disruption of normal brain development leads to an array of developmental disorders. One of the most common of these is mental retardation, the prime cause of which is prenatal exposure to alcohol. As chapters in this book show, alcohol has direct effects on the developing neural system and it affects genetic regulation. Another common neurotoxin is nicotine, and it is discussed in this book for three reasons: (1) the number of adolescents who smoke cigarettes is rising in some populations; (2) prenatal exposure to nicotine affects neurotransmitter systems that are critical for normal brain development and cognition; and (3) prenatal exposure to nicotine is often accompanied by prenatal exposure to alcohol.LThe mature brain is the culmination of an orderly sequence of the basic ontogenetic processes--cell proliferation, migration, differentiation, and death. Neural stem cells and progenitors proliferate in discrete sites; then, young neurons migrate long distances to their residences where they form neural networks. During this sequence many immature cells die, presumably eliminating unsuitable or non-competitive cells. Each process is regulated by genetic and environmental factors. When this regulation goes awry, a dysmorphic and dysfunctional brain results. Though this can be tragic in clinical settings, in experimental contexts it provides keen insight into normal brain development.LThe book is divided into three parts. The first describes neural ontogeny in the normal brain. The second and third deal with the consequences of early exposure to alcohol and nicotine. Though there are similarities in the effects of these two toxins, there are also intriguing differences. The commonalities reflect the plasticity and resilience of the developing brain while the differences point to the targeted effects of the two toxins. Exploring these effects brings a richer appreciation of brain development. The book will be of interest to neuroscientists, developmental biologists, teratologists, pharmacologists, toxicologists, neurologists, neuropsychologists, and to their students and trainees.
